微小RNA-155在M1型、M2型巨噬细胞及肿瘤相关巨噬细胞中的表达差异 被引量:2

Expression Difference of Micro RNA-155 among Macrophages M1, M2, and Tumor-associated Macrophages

摘要 目的探讨微小RNA-155(mi R-155)在巨噬细胞M1型、M2型及肿瘤相关巨噬细胞(TAMs)中的表达差异。方法将单核细胞系(THP-1)细胞诱导为巨噬细胞M1型、M2型及TAMs,应用流式细胞仪检测表型后,采用荧光染料法实时定量PCR(SYBR Green q RT-PCR)方法分别检测THP-1、巨噬细胞M1型、M2型及TAMs中mi R-155的表达情况。结果 M1型巨噬细胞中mi R-155的表达量为6.580±0.637,M2型巨噬细胞中的表达量为1.83±0.337,TAMs细胞中的表达量为1.60±0.233。单因素方差分析结果显示组间差异有统计学意义(F=1.238,P=0.000)。其中M2型(P=0.000)及TAMs(P=0.000)巨噬细胞中mi R-155的表达量显著低于M1型;TAMs的表型与巨噬细胞M2型的表型相似,TAMs与M2型巨噬细胞中mi R-155的表达差异无统计学意义(P=0.546)。结论 mi R-155在巨噬细胞M1型、M2型中的差异性表达可能与其分化和/或细胞功能相关。TAMs的表型特征可能与巨噬细胞M2型类似,同时可能具有与巨噬细胞M2型相同的细胞功能。 Objective To investigate the expressions of micro RNA-155(mi R-155) in different phenotypes of activated macrophages. Methods The THP-1 cells underwent polarized activation into M1, M2 or tumor-associated macrophages(TAMs), and the phenotypes were confirmed by flow cytometry. The mi R-155 expression was determined by q Rt-PCR in M1 macrophages, M2 macrophages and TAMs. Results The mi R-155 expression significantly decreased in the M2 macrophages(1.83±0.337, P=0.000), TAMs(1.60±0.233, P=0.000) compared with the M1(6.580±0.637). The phenotype of TAMs was similar to M2. There was no statistically significant difference between TAMs and M2 macrophages in the expression of mi R-155(P=0.546). Conclusion Different expressions of mi R-155 in macrophages M1-type and M2-type may be associated with the differentiation or their cellular functions. The phenotypic characteristics TAMs may transform to macrophages to M2-type. And they may have the same functions.
