党参多糖类对荷瘤小鼠免疫应答及抑瘤作用研究 被引量:48

Effect of wen codonopsis pilosula and baitiao codonopsis pilosula-polysaccharide on the S180 tumor and the blood cytokines in mice

摘要 目的探讨纹党参多糖(wen codonopsis pilosula polysaccharide,WCPP)及白条党多糖(baitiao codonopsis pilosula polysaccharide,BTCPP)的抗肿瘤作用机制及其对外周血细胞因子的影响。方法小鼠100只,简单随机分组法分成空白对照组、阴性对照组(模型对照组)、阳性对照组(CTX组)、WCPP组和BTCPP组,每组20只。除空白对照组外,其余各组均接种S180瘤细胞。空白对照组和荷瘤模型组给予生理盐水,CTX组给予环磷酰胺,WCPP和BTCPP组分别连续用药20d,比较各组肿瘤生长情况,并计算抑瘤率和脾脏以及胸腺指数。MTT法测定淋巴细胞增殖刺激指数,乳酸脱氢酶释放法测定NK细胞活度,ELISA试剂盒检测白细胞介素2(interleukin-2,IL-2)、IL-6、IL-1β、IL-4、干扰素γ(interferon gamma,INF-γ)和肿瘤坏死因子α(tumor necrosis factor-alpha TNF-α)等细胞因子。结果 WCPP抑瘤率为49.22%,BTCPP为47.28%,对小鼠移植性肿瘤具有明显的抑制作用。WCPP和BTCPP可显著改善荷瘤小鼠胸腺指数及脾脏指数,增强荷瘤小鼠NK细胞活度及淋巴细胞增殖指数,与荷瘤模型组比较P值均<0.05。在NK细胞活度和淋巴细胞增殖刺激指数的影响方面,WCPP组与BTCPP组差异有统计学意义,P值分别为0.032和0.001。WCPP和BTCPP也能显著增加荷瘤小鼠血清细胞因子IL-2、IL-1β、IL-6、TNF-α和INF-γ的含量,P值均<0.05;同时降低血清IL-4的水平,与荷瘤模型组比较差异有统计学意义,P<0.05;且WCPP组和BTCPP组血清IL-2、IL-4水平差异有统计学意义,P值均<0.05。结论 WCPP和BTCPP对小鼠移植性肿瘤具有显著的抑制效应;在对NK细胞活度和淋巴细胞增殖刺激指数及荷瘤小鼠IL-4和IL-2水平的影响方面,WCPP的作用明显优于BTCPP。 OBJECTIVE To explore the mechanism of wen codonopsis pilosula polysaccharide(WCPP) and baitiao codonopsis pilosula polysaccharide(BTCPP) on mouse's transplantable tumor and CK in blood, which could provide theo- retical evidence for the development of Radix codonopsis pilosula. METHODS Totally 100 mice were divided into 5 groups randomly and each group 20, including the blank-control group(BC group), the tumor-burdened model group (Model group), WCPP group, BTCPP group and Cytoxan group (CTX group). All the mice were inoculated with $180 tumor cells except for the blank control group. The BC and Model group were administrated saline. WCPP, BTCPP and CTX group were treated with WCPP, BTCPP and CTX for 20 day respectively. The tumor growth was compared in all groups. The inhibitory rate of tumor, spleen and thymus index were observed. The stimulate lymphocyte proliferation in dex was determined by MTT method. NK cell activity was analysis by lactate dehydrogenase release method. IL-2, IL-6, IL-1β, IL-4, INF-gamma and TNF alpha was tested with ELISA kits. RESULTS WCPP and BTCPP could effectively inhibit the mouse' s transplantable tumor. The inhibitory rate of tumor was 49. 22% and 47. 28% respectively. The WCPP and BTCPP could improve the thymus index, the spleen index, NK cell activity and the ability of lymphocyte pro- liferation of mice burdened S180 tumor. There was significant difference between WCPP group or BTCPP group and mod el group (P〈0.05). There were significant differences in the NK cells activity and lymphocyte proliferation stimulus in dex between the WCPP group and BTCPP group (P values were 0. 032 and 0. 001, respectively). All of the data about the IL-2,IL-1β, IL-6, IL-4, TNF-α, INF-γ of groups treated with WCPP and BTCPP were different compared with the model group. When tumor- burdened mice treated with WCPP and BTCPP, IL-2, IL-1β, IL-6, TNFα and INF-γ were in- creased, while IL-4 was decreased. The level of IL-2 and IL-4 in serum treated with WCPP and BTCPP was significantly different(P〈0.05). CONCLUSIONS WCPP and BTCPP have an obvious effect on the mouse's transplantable tumor. WCPP was better than BTCPP for the influence on NK cell activity, the ability of lymphocyte proliferation, IL-4 and IL-2 level in serum.
