辣椒根尖预处理及植株倍性鉴定 被引量:7

摘要 采用0.1%秋水仙素、0.002 mol/L 8-羟基喹啉和冰水3种试剂对辣椒辛香8号花药培养再生植株根尖进行预处理,研究不同预处理对根尖细胞中期分裂相的影响效果,并对辣椒花药培养再生植株进行倍性鉴定,以期获得辣椒染色体计数的最佳预处理。结果表明:3种预处理中,利用0.1%秋水仙素对辣椒根尖预处理3 h,细胞的中期分裂指数最高,达56.6‰,而且染色体分散度好,染色体分裂相多,比较适合染色体计数;经染色体计数鉴定得出5株花药培养再生植株中有4株双倍体和1株单倍体,可用于下一步育种实践中。 Pepper Xiangxin8 root tip were pretreated with three reagents including 0.1% colchicine,0.002 mol/L 8-hydroxyquinoline and ice water to study the effect of different pretreatment on root tip cells in metaphase of pepper,and the ploidy was identified of another culture derived pepper plants to obtain well pretreatment for chromosome counting. The results showed that metaphase mitotic index was as high as 56.6% in the density of 0.1% colchicines and 3 hours treatment,which the chromosomes spreader well,more split phases,and were suitable for counting. 4 haploid and 1 diploid were identified in 5 regenerated plants by chromosome counting. These could be used for breeding.
