细胞因子信号转导抑制因子3在早产胎盘中的表达及临床意义

Expression of SOCS3 and Its Significance in Preterm Placenta

目的 探索细胞因子信号转导抑制因子3在早产胎盘中的表达及临床意义。方法 选取2013年4-10月解放军第三〇九医院收治的20例早产妇女作为实验组,20例足月妊娠妇女作为对照组,收集两组胎盘组织。应用Western blot法检测两组胎盘组织中SOCS3分子的表达水平,采用免疫组织化学法检测两组细胞因子白细胞介素（IL）10、肿瘤坏死因子（TNF）α的表达水平。比较2种因子在两组胎盘中的差异表达情况。结果 与对照组相比,实验组胎盘组织中SOCS3阳性表达量明显增高（1.09±0.29比0.81±0.22,P〈0.05）,IL-10阳性表达率明显高于对照组（85.0%比45.0%;P〈0.05）,而实验组TNF-α表达阳性率则明显降低（25.0%比70.0%,P〈0.05）。结论 SOCS3及IL-10在早产胎盘中表达较高,相反TNF-α表达降低,其是可能SOCS3是通过对Th1/Th2的平衡调节,导致了早产的发生重要原因之一。

Objective To investigate the expression of suppressor of cytokine signaling（ SOCS3） and its significance in preterm placenta. Methods Retrospective analysis was made on 20 patients（ observation group） who had spontaneous preterm labor and 20 pregnant women（ control group） with normal delivery during the same period,the patients were from PLA 309 th Hospital during Apr. 2013 and Oct. 2013. The expression of SOCS3 in placenta were examined by Western blot,the expression of interleukin（ IL）-10 and tumor mecrosis factor α（ TNF-α） was detected by immunohistochemical method. The differences of expression of SOCS3,IL-10,TNF-α were compared. Results Compared with normal delivery,the expression of SOCS3 and IL-10 were significantly increased in placenta with preterm birth（ SOCS3： 1. 09 ± 0. 29 vs 0. 81 ±0. 22,P〈0. 05; IL-10： 85. 0% vs 45. 0%,P〈0. 05）,and expression of TNF-α was significantly decreased in placenta with preterm birth（ 25. 0% vs 70. 0%,P〈0. 05）. Conclusion The expression of SOCS3 and IL-10 were increased and TNF-α was decreased in placenta with preterm birth,and SOCS3 may influence the Th1 / Th2 balance and is one of the important causes leading to preterm birth.
