摘要
目的探讨熊果酸对蛛网膜下腔出血(Subarachnoid hemorrhage,SAH)后早期脑损伤的保护作用。方法 48只成年雄性SD大鼠分成4组(假手术组、模型组、低剂量给药组、高剂量给药组),每组12只。采用颈内动脉穿刺法建立大鼠SAH模型,造模后24 h进行神经功能缺陷评分;造模后72 h检测脑含水量、脑血管通透性;造模后72 h检测血清中丙二醛(Malondialdehyde,MDA)、超氧化物岐化酶(Superoxide dismutase,SOD)和谷胱甘肽过氧化物酶(Glutathione peroxidase,GSH-Px)活性。结果熊果酸给药可有效改善SAH大鼠的神经功能缺损(P<0.05);降低脑含水量和脑血管通透性(P<0.05);降低血清MDA含量(P<0.05);增强SOD和GSH-Px活性(P<0.05)。结论熊果酸对SAH后早期脑损伤有显著保护作用,其机制可能与提高脑组织抗氧化能力有关。
Objective To explore the effects of ursolic acid ( UA) on early brain injury following subarach-noid hemorrhage ( SAH) . Methods 48 male SD rats were divided into vehicle-treated sham,vehicle-treated SAH,and UA-treated SAH groups. The endovascular puncture model was used to induce SAH. Neurological score was evaluated at 24 h after SAH, brain water content and blood-brain barrier ( BBB ) permeability were measured at 72 h after SAH. The levels of serum superoxide dismutase (SOD),malondialdehyde(MDA) and glutathione peroxidase (GSH-Px) were determined by diagnostic kit. Results UA administration could significantly improve neurological scores (P〈0.05),reduce brain water content and BBB permeability (P〈0. 05),decrease the serum level of MDA (P〈0.05),increase the level of SOD and GSH-Px (P〈0. 05). Conclusion UA could protect against EBI following SAH and its mechanism maybe partly attribute to its antioxidant property.
