摘要
通过紫外光谱研究了四苯基卟啉的紫外可见光谱性质,H2TPP有较强的Soret带和较弱的4个Q带。并通过荧光光谱研究了H2TPP与BSA猝灭作用,在不同的温度(295.15,310.15 K)下,通过模型方程等计算了该过程,猝灭速率常数6.71×105,6.21×105L/(mol·L·S),结合常数5.55×105,7.53×105L/mol结合位点1.058,0.949,判定该过程为静态猝灭。并通过热力学常数的变化,△G、△H、△S<0,说明该过程是自发进行,主要作用力是氢键和范德华力,通过同步荧光光谱,判断该结合作用主要在BSA的色氨酸残基位置。
The characteristics of spectrum was studied by UV-Visible,it indicated that was the strong Soret band and four Q bands existed. The quenching of BSA and H2 TPP reaction was studied by fluorescence spectra. The binding were measured at different temperatures (295. 15,310. 15 K). The equilibrium con-stant was 6.71 ×105,6.21 ×105 L/(mol·L·S),the binding constant was 5.55 ×105,7.53 ×105 L/mol,the molar ratio was 1. 058,0. 949. The experiment showed that the sort of quenching was determined as static quenching. According to the thermodynamic parameters,△G,△H and△S〈0,the main sorts of bind-ing force could be judged as hydrophobic and van der Waals’ force. It was concluded that the mainly inter-act with BSA locating near the Trp residues,by using the synchronous fluorescence spectra.
