摘要
对"建莲"莲子心中3种生物碱类成分进行含量测定,并建立HPLC指纹图谱分析方法,用于"建莲"莲子心的化学质量评价。采用Thermo C18(4.6 mm×250 mm,5μm)为色谱柱,乙腈-十二烷基磺酸钠溶液-冰醋酸(56∶43∶1)为流动相,流速1.0 m L·min-1,检测波长282 nm,柱温35℃,对"建莲"莲子心中3种生物碱类成分进行含量测定。采用Agilent ZORBAX SBAq C18(4.6 mm×250 mm,5μm)为色谱柱,甲醇-水为流动相,流速0.8 m L·min-1,检测波长282 nm,柱温35℃,建立"建莲"莲子心的HPLC指纹图谱,并应用相似度评价和聚类分析的方法对12批"建莲"莲子心样品的HPLC指纹图谱数据进行分析。结果显示11批"建莲"莲子心样品在化学成分上具有较好的一致性。该方法较好地反映了"建莲"莲子心中的化学成分信息,且操作简单,重复性好,为进行"建莲"莲子心的化学质量评价提供了参考。
This study is to determine the content of three alkaloids and establish the HPLC fingerprint of "Jianlian" Nelumbinis Plumula. The HPLC method of content determination was as follows : Thermo C18 (4. 6 mm × 250 mm ,5 μm) was conducted with ace- tonitrile-sodium dodecyl sulfonate solution-acetic acid (56: 43:1 ) at a flow rate of 1.0 mL · min-1. The monitoring wavelength was set at 282 nm and the column temperature was 35 ℃. The method of HPLC fingerprint was as follows: Agilent ZORBAX SB-Aq C18 (4. 6 mm ×250 mm, 5μm) was conducted with gradient elution of methanol and water at a flow rate of 0. 8 mL . min-1, the monitoring wavelength was set at 282 nm and the column temperature was 35 ℃. Similarities evaluation and hierarchical clustering analysis were applied to demonstrate the variability of 12 batches of "Jianlian" Nelumbinis Plumula samples. The results demonstrated that 11 bat- ches showed good similarity on chemical constituents. The method could well display the chemical information of "Jianlian" Nelumbinis i Plumula. It was simple, reliable and could be used for the chemical quality control of "Jianlian" Nelumbinis Plumula.
