摘要
目的通过观察RKI-1447对人卵巢癌CAOV-3细胞生长及凋亡的影响,探讨其在体外的抗肿瘤效应。方法体外培养CAOV-3细胞,以不同浓度的RKI-1447为实验组,并设置对照组。CCK8法测细胞增殖抑制效应,光学显微镜观察药物对细胞形态学的影响,流式细胞术检测CAOV-3细胞的凋亡情况。结果 CAOV-3细胞24 h增殖抑制效应有剂量依赖性,RKI-1447浓度为10μmol/L时其抑制效应与其他浓度相比,差异有统计学意义(P<0.05),并可诱导CAOV-3细胞凋亡。结论 RKI-1447能够抑制CAOV-3细胞的生长,其增殖抑制效应具有剂量依赖性,同时可以诱导细胞凋亡。
Objective To observe the effect of RKI-1447 on cell growth and apoptosis of ovarian cancer CAOV-3 cells, in order to investigate the anti-tumor effect of RKI-1447 in vitro. Methods CAOV-3 cells were cultured in vitro. In experimental group, the cells were treated by different concentrations of RKI- 1447, while control group was also assigned. Inhibitory effect on cell proliferation was deter- mined using CCKS, and the effect on cell morphology was observed under light microscope, and apoptosis of CAOV-3 ceils was identified u- sing flow cytometry. Results Inhibitory effect on CAOV-3 cell proliferation in 24 hours was in a dose-dependent manner. The inhibitory effect of RKI-1447 at 10 μmol/L had significant difference compared with other concentrations (P〈0.05), which could induce CAOV-3 cells apoptosis. Conclusion RKI-1447 can inhibit CAOV-3 cell growth in a dose-dependent manner and induce cell apoptosis.
