摘要
目的:探讨不同灸温对佐剂性关节炎大鼠血清炎症因子的影响及瞬时受体电位香草酸亚型1(TRPV1)的作用机制。方法:将40只SD大鼠随机分为空白组、模型组、38℃艾灸组、45℃艾灸组、CPZ+45℃组,每组8只,以弗氏完全佐剂制备佐剂性关节炎模型。后3组于"肾俞"、"后三里"给予每天每穴10 min治疗,控制穴区温度为(38±1)℃或(45±1)℃。CPZ+45℃组先在穴区涂抹TRPV1阻断剂辣椒平CPZ溶液,每10 min涂1次,30 min后再予45℃艾灸治疗。治疗5 d后测定肿瘤坏死因子(TNF)-α、白介素(IL)-1β的浓度。结果:模型组TNF-α、IL-1β较空白组显著升高。38℃组TNF-α较模组型降低,而IL-1β无统计学意义,45℃组TNF-α、IL-1β较模型组显著降低。45℃组TNF-α、IL-1β较38℃组明显降低。CPZ+45℃组TNF-α、IL-1β较45℃组明显升高。结论:灸法具有良好的抗炎作用,灸温的高低直接影响治疗疗效,TRPV1在感知灼痛温度(>43℃)后,参与了艾灸抗炎效应。
Objective: To explore the influence of different moxibustion temperatures on adjuvant arthritis rats serum inflammatory cytokines and transient receptor potential vanilloid 1 TRPV1 mechanism. Method: The 40 SD rats were randomly divided into a blank group, a model group, 38℃ moxibustion group, 45℃ moxibustion group and CPZ + 45℃ group. Using the freund's complete adjuvant (FCA) established the model of adjuvant arthritis (AA) rats. Moxibustion was applied at the last three groups at "Shenshu" (BL23)and "Housanli" (ST36) for 10 min per acupoint per day,at the same time controlling the temperature at (38 + 1 ) ℃ or (45 + 1 ) ℃. CPZ +45℃ group daubed the TRPV1 blocker CPZ solution at acupoints, daubing once every 10 minutes, after thirty minutes moxibustion was applied controlling the temperature at (45±1 )℃ simultaneously. After continuous five days treatment, serum was acquired. Enzyme-linked immune sorbent assay (ELISA) method was adapted to measure the concentration of tumor necrosis factor TNF-α and interleukin IL-1β. Results: The TNF-α and IL-1α in the model group were significantly higher than those in the blank group. The TNF-α of the 38℃ group was lower than model group, but the comparison of IL-1β without statistical significance,those of 45 ℃ group were lower than those of model group. The TNF-α and IL-1β of the 45℃ group were significantly lower than those of the 38℃ group. The TNF-α and IL-1β of the CPZ + 45℃ group were significantly higher than those of the 45℃ group. Conclusion:Moxibustion has good anti-inflammatory effects, The temperature of high and low directly affects treatment efficacy, TRPV1 in perception of hot temperature above 43℃ participates in the anti-inflammatory effect of moxibustion.
