艾叶燃烟自由基的细胞毒性作用研究 被引量:4

摘要 目的:通过自由基再现技术和体外细胞毒性试验研究艾烟自由基的细胞毒性作用。方法:采用对苯二酚在碱性条件下再现产生自由基,选用L6、HepG2和A549与其作用,通过MTT法、荧光染色观察法判定待检样品是否对细胞有毒性作用,并进行流式细胞术分析。结果:在12.5~200μmol/L浓度下,相同浓度对苯二酚在不同的pH条件下对细胞毒性作用的强弱不同。对苯二酚在偏酸性条件下,半数抑制浓度(IC50)值大,细胞毒性小;对苯二酚在偏碱性条件下,IC50值小,细胞毒性大,且碱性组较酸性组毒性增加明显,P〈0.05或P〈0.01。结论:对苯二酚在碱性条件下的细胞毒性作用远远强于其在偏酸性条件下,参与其细胞毒性的物质包括半醌自由基、对苯二酚及对苯二酚聚合物。研究艾烟的毒性作用不仅要考虑艾烟中的稳定物质,还要考虑到艾烟中的不稳定物质——自由基。 Objective:The study aims at finding out the cytotoxic effect of the free radical in mugwort fumigation through free radical reappearance technology and in vitro cytotoxicity - test. Method: Free radical can be detected in mugwort fumigation and it can reappearance by hydroquinone in alkaline conditions. We carried out cytotoxicity tests by MTT method and fluorescent staining method to determine whether the sample has toxic effect on ceils, which are L6, HepG2 and A549. Results :The same concentration of hydroquinone in different pH conditions has different cell toxicity interaction. Hydroquinone in acidic conditions ,we got large ICs0 value, which means it has low cytotoxicity;hydroquinone in alkaline conditions,we got small IC50 value, which means it has high cell toxicity.
