半夏泻心汤抗大鼠应激性胃黏膜损伤及对炎性相关因子含量的影响 被引量:6

Effects of Banxia Xiexin Decoction against Rats Stress-Induced Gastric Mucosal Injury and Content of Inflammatory Related Factor

摘要 目的:观察预先给予辛开苦降的代表方半夏泻心汤对水浸束缚应激大鼠血清IL-1β、IL-8、TNF-α、IL-10含量的影响,探讨辛开苦降法抗应激性胃黏膜损伤的作用机制。方法:SD大鼠随机分为空白组、模型组、中药组(半夏泻心汤)、西药组(奥美拉唑)。每组动物相应药物灌胃3 d。除空白组外,其他大鼠水浸束缚应激(WRS)造模。计算大鼠胃黏膜损伤指数(UI),HE染色光镜下观察大鼠胃黏膜病理改变,酶联免疫法检测各组大鼠血清IL-1β、IL-8、TNF-α、IL-10的含量。结果:与空白组比较,模型组大鼠胃黏膜损伤指数明显升高,病理改变明显,促炎因子IL-1β、IL-8、TNF-α含量显著升高(P<0.01),抑炎因子IL-10含量显著降低(P<0.01)。与模型组比较,预先给予中药处理UI明显降低(P<0.01),促炎因子IL-1β、IL-8、TNF-α含量显著降低(P<0.01),抑炎因子IL-10含量显著升高(P<0.01)。结论:辛开苦降的半夏泻心汤可显著对抗应激性胃黏膜损伤,其机制可能是通过升高抑炎因子IL-10、降低促炎因子IL-1β、IL-8、TNF-α的含量实现的。 Objective :To observe the effect of Xinkai Kujiang representative formula Banxia Xiexin Decoction on restraint stress rats serum IL - 1β, IL - 8, TNF - α and IL - 10 contents and explore the mechanism of Xinkai Kujiang method against stress induced gastric mucosal injury. Methods:SD rats were randomly divided into four groups:blank group, model group, Chinese medicine group ( Banxia Xiexin Decoction), western medicine group (Omeprazole). Each animal was given corresponding drugs by garage for 3 days. Except the blank group, the other rats were made animal modei by water immersion restraint stress (WRS). Calculate the index of gastric mucosal injury in rats and histopathological changes of gastric mucosa in rats under light microscope HE staining. Detect the contents of IL - 1β, IL - 8, TNF - α and IL - 10 in serum in rats by enzyme -linked immunosorbent assay. Results: Compared with the blank group,gastric muco- sal injury index in rats of model group increased obviously and pathological changes were obvious. Proinflammatory factors IL-1β,IL-8 and TNF-α contents were significantly increased (P 〈0.01 ). Anti -inflammatory factor IL- 10 was significantly lower(P 〈 0.01 ). Compared with the model group, UI significantly reduced after pre - intervention of TCM ( P 〈 0.01 ). Proinflammatory factors IL - 1β, IL - 8 and TNF - α were significantly lower ( P 〈 0.01 ). Anti inflammato- ry factor IL - 10 content was significantly increased ( P 〈 0.01 ). Conclusions : Banxia Xiexin Decoction of Xinkai Kujiang Method can be obviously against stress - induced gastric mucosal injury. The mechanism is probably actualized through increasing anti inflammatory factor IL - 10 and reducing the contents of implementation of proinflammatory cytokines IL - 1β,IL-8 and TNF -α.
