芹菜素对急性大鼠脑缺血再灌注后脑组织NCAM表达的影响 被引量:2

Effects of Apigenin on NCAM Expression in Brain Tissue after Acute Focal Cerebral Ischemia-Reperfusion Rats

摘要 目的:探讨NCAM在大鼠脑缺血再灌注后不同时间的表达变化及芹菜素对其表达的影响。方法:采用改良线栓法建立大鼠脑缺血再灌注后不同时间损伤模型。动物随机分为假手术组(S)、模型组(M)、芹菜素组(A)和地塞米松组(D)。各手术组按再灌注时间不同又分为再灌注6 h组、24 h组、72 h组、7 d组,各组动物麻醉清醒后均给予神经行为学评分,每组1只大鼠TTC染色观察脑梗死灶、6只免疫组化法测定NCAM的表达。结果在A72h组神经行为学评分低于M72h(P<0.05);脑组织切片TTC染色显示各手术组均出现白色梗死灶;正常脑组织NCAM少量表达,缺血再灌注后表达增加,于72 h达高峰,A组能显著提高NCAM的表达,各时间点A和M组比较差异显著(P<0.05)。结论:芹菜素可增加脑缺血再灌注后NCAM的表达,提示芹菜素可能通过NCMA达到促进脑组织损伤后神经细胞修复和再生的作用。 Objective :To investigate the effects of apigenin (APG) on expression of NCAM of focal cerebral ischemia - reperfusion rats. Methods : Cerebral ischemia was induced by middle cerebral artery occlusion. The rats were randomly divided into four groups ineluding sham - operated, model, APG and Dexamethasone groups. All the operated groups were divided into different groups:reperfusion for 6 h,24 h,72 h and 7 d groups. Neurological behavior was assessed by five score methods. After anesthesia,each groups animals were given neurological behavior scores. The brain slices of one rat in each group were observed by TTC stainning and the expression of NCAM in another six brain slices were measured by immunohistochemistry. Results : Neurological behavior scores of the rats in A72h group was significant lower than those in ischemia M72h (P 〈 0.05). Typical cortical infarct lesions in model group were found by TTC stainning. Normal brain tissue NCAM expression was less. The expression of NCAM was enhanced immediately after ischemia and reached to peak at 72 h after ischemia respectively. Apigenin could significantly increase the expressions of NCAM than ones at the corresponding time points in group M respectively ( P 〈 0.05 ). Conclusion : Apigenin could increase NCAM expression in transient focal cerebral ischemia and reperfusion rats. They might repair ischemia brain tissue and promote nerve cell repair and regeneration through NCAM.
