摘要
目的:观察回神颗粒对老年痴呆模型小鼠P13K/Akt信号传导通路的影响。方法:老年痴呆模型APPswe/'PS1△E9小鼠40只,随机分为2组,即模型组和回神颗粒组;同背景同月龄的C57B6J小鼠20只,随机分为两组,即正常组和正常加药组,回神颗粒组和正常加药组灌服回神颗粒3个月,通过Western Bloting方法检测小鼠脑组织中Akt、phospho-Akt(pAkt)、CREB、phospho-CREB(P-CREB)、Bcl-2和Bax的蛋白表达。结果:与正常组相比,模型组的p-Akt、p-CREB、Bcl-2的表达水平明显减低,Bax表达水平明显增加(P<0.05)。与模型组相比,回神颗粒组小鼠的p-Akt、p-CREB、Bcl-2的表达水平明显增加,Bax表达水平明显降低(P<0.05)。结论:回神颗粒改善APPswe/PS1△E9双转基因小鼠认知功能障碍的作用可能是通过调控P13K/Akt信号通路起作用的。
Objective : To observe the effect of huishen granules on the PI3 K/Akt signalling of model mice with senile dementia. Method: g0 model APPswe/PS1AE9 mice with senile dementia were randomly divided into two groups, i.e. model group and huishen granules group; 20 C57B6J mice with the same background and age were randomly divided into two groups, i.e. normal group and normal medication group. Huishen granules group and normal medication group were given huishen granules for three months, The protein expression of Akt,phospho-Akt (p-Akt) ,CREB,phospho-CREB (p-CREB),Bcl-2 and Bax in brain tissues of mice was determined by Western bloting. Results: Compared with normal group, the expressed level of p-Akt,p-CREB,Bcl-2 in model group was significantly decreased, and the expressed level of Bax rose significantly ( P 〈 0.05 ) ; compared with model group, the expressed level of p-Akt,p-CREB,Bcl-2 in huishen granules group rose significantly, and the expressed level of Bax decreased significantly(P 〈0.05 ). Conclusion: Huishen granules play a part in improving cognitive dysfunction of APPswe/PS1 AE9 double-transgenic mice by regulating PI3K/Akt signalling.
