摘要
利用农杆菌介导法将组成型表达启动子CaMV35S驱动的正义和反义无机焦磷酸酶(PPase)基因导入到马铃薯栽培品种‘费乌瑞它’的试管薯中,通过PCR检测证明PPnse基因已转入马铃薯基因组中.对转基因株系及对照植株块茎的休眠特性的测定表明,分别在4℃低温和25℃室温贮藏条件下的转反义基因马铃薯株系块茎的休眠期较对照株系均延长了2~3周;转正义基因马铃薯株系块茎的休眠期较对照株系均提前了2~3周;且在4℃低温贮藏条件下的转基因马铃薯株系及对照块茎的休眠期比25℃条件贮藏的长2~3周,为进一步利用PPase基因改良马铃薯块茎休眠特性提供科学依据.
Microtubers of potato cv. 'Favorita' were used as explants in this experiment, the plant ex- pression vectors of sense and antisense PPase gene were driven by the constitutive expression promoter CaMV 35S,which were constructed and transferred to potato by Agrobacterium-mediated transformation method. And PCR assay confirmed that both sense and antisense PPase gene were integrated into potato genome. Tuber dormancy characteristic of the transgenic plants stored at 4 ℃ and 25 ℃ showed that period of dormancy of the transgenic antisense PPase gene plants were prolonged 2 to 3 weeks compared to the control,while the dormancy period of the transgenic sense PPase gene plants were shortened 2 to 3 weeks. The results provide the theoretical evidence for further improving potato tuber dormancy characters using PPase gene.
