肿瘤抑制因子p96蛋白在小鼠内耳毛细胞的表达

摘要 目的研究肿瘤抑制因子p96蛋白在昆明小鼠内耳毛细胞的定位表达特点。方法采用间接免疫荧光法标记耳蜗基底膜并铺片,激光共聚焦显微镜分层扫描观察p96蛋白的表达定位。结果内、外毛细胞中均可观察到p96蛋白荧光标记,且该蛋白在毛细胞胞质中的表达并不均匀。在内毛细胞胞质中,细胞顶部表皮板及附近胞质区有类圆形的P96蛋白高表达区。结论肿瘤抑制因子p96蛋白表达于内、外毛细胞,在内毛细胞顶区有着显著的局部高表达特征。结合该蛋白的生理功能,推测内毛细胞顶区可能是囊泡内吞、物质交换的一个重要亚细胞部位。 Objective To investigate the expression of tumour suppressor, p96, in the cochlear hair cells in Kunming mice. Methods The basilar membrane of mice cochlea was immuno-double labeled with antibody and Dapi. The distribution of p96 was detected by immunofluorescence staining and laser scanning confocal microscope. Results P96 was expressed in the inner and outer hair ceils of cochlea and was especially strong in the inner hair cells at the apical area. This area ought to be the cuticular plate and the nearby cytoplasma. Conclusions The expression of tumor suppressor, p96, displayed a prominent polarised characteristics which may be similar to the expression of polarised epithelial cells. The cuticular plate and the nearby cytoplasma of inner hair cells maybe a key subeeUular site related to the differentiation and metabolism of cells.
