硫化氢对内皮素诱导的大鼠心肌肥厚的作用研究

摘要 目的探讨硫化氢(H2S)对内皮素诱导的大鼠心肌肥厚的影响。方法取64只成年雄性大鼠,体质量180-200 g,随机分为4组:正常组10只、假手术组10只、内皮素诱导心肌肥厚模型组22只、内皮素诱导心肌肥厚+H2S(56μmol/kg)治疗组22只。4周之后检测左心室质量指数、心肌细胞肥大形态学指标、心肌间质胶原容积分数以及心肌羟脯氨酸含量等各项心肌肥厚指标。结果与单纯心肌肥厚组相比,H2S治疗组左心室质量下降21.4%,左心室质量指数下降5.97%,心肌细胞最短径下降12.5%,心肌细胞横断面面积下降10.8%,心肌间质胶原容积分数下降22.3%,心肌羟脯氨酸含量下降31.3%(P均〈0.05)。结论 H2S可降低内皮素诱导的心肌肥厚大鼠左心室质量指数,改善心肌细胞肥大,降低心肌胶原蛋白水平以及心肌纤维化,为治疗心肌肥厚提供思路。 Objective To evaluate the effect of hydrogen sulfide molecules on cardiac hypertrophy induced by endothelin in rats. Methods Sixty-four adult male rats,weighing 180-200 g,were divided into 4 groups randomly with10 rats in normal group,10 rats in sham group,22 rats in model group( cardiac hypertrophy model induced by endothelin),22 rats in treatment group( cardiac hypertrophy rats induced by endothelin treated with H2S). After four weeks,left ventricular mass fraction,morphological index of myocardial hypertrophy,myocardial interstitial collagen volume fraction and myocardial hydroxyproline content and other indicators of cardiac hypertrophy were detected. Results Compared with the model group,left ventricular mass reduced by 21. 4%,left ventricular mass index reduced by5. 97%,cardiomyocytes shortest diameter reduced by 12. 5%, myocardium cell cross section reduced by 10. 8%,myocardial interstitial collagen volume fraction reduced by 22. 3%,myocardial hydroxyproline content reduced by 31. 3%in treatment group( P 〈 0. 05). Conclusions H2 S can lower left ventricular mass fraction,improve myocardial hypertrophy,reduce myocardial collagen levels and decrease myocardial fibrosis. It can provide ideas for new drug research and development on myocardial hypertrophy.
