摘要
通过改进普通CTAB法,获得了适用于太湖洞庭地区收集的红桔资源基因组DNA提取方法,结果表明得到的DNA质量较高,符合ISSR-PCR扩增要求.利用ISSR分子标记对26份红桔资源进行亲缘关系分析,8个引物共扩增出105条条带,其中91条为多态性条带,多态率为86.7%.利用NTsys 2.10e软件进行UPGMA聚类分析,26份资源的遗传相似性系数在0.62~0.86之间,0.73处所有资源分成4类.遗传相似性结果将为红桔种质资源分类和育种提供理论依据.
Common CTAB method was improved to extract genomic DNA of Citrus reticulata. The results showed that high quality DNA for ISSR'-PCR analysis was produced by this method. 26 Citrus samples were examined with ISSR technique to construct DNA fingerprinting and evaluate their genetic relationship. A total of 105 bands were obtained through 8 primers, of which 91,86.7% , v/ere polymorphic. The genetic distance was analyzed, and the genetic similarity coefficient were be- tween 0.62 and 0.86. A tree diagram was constructed with NTsys 2.10e software , which 26 samples could be clustered into 4 groups at the similarity coefficient level of 0.73. The result of similarity coefficient could be used for classification and breeding of the Citrus resources.
