摘要
目的探讨姜黄素对大鼠脑缺血损伤时脑组织磷脂酰肌醇3激酶(PI3K)/丝氨酸/苏氨酸蛋白激酶(AKT)信号转导通路、caspase-3表达的影响及可能机制。方法健康SD雄性大鼠随机分为四组:假手术组、模型组、姜黄素A组和姜黄素B组,每组大鼠18只,免疫组化法检测PI3K、AKT蛋白表达,ELISA法检测caspase-3的含量。干湿重法检测脑组织水含量,甲酰胺法检测血脑屏障通透性。结果模型组比较,姜黄素组PI3K、AKT蛋白均明显增高(P<0.05,P<0.01),脑组织含水量、caspase-3及伊文思蓝含量显著降低(P<0.05,P<0.01,P<0.01)。结论姜黄素可能通过激活PI3K/AKT信号通路,下调caspase-3的表达,从而减轻脑缺血性损伤。
Objective To expIore the effects of curcumin on cerebraI ischemia in rat brain PI3K/AKT signaI transduction pathway and the possibIe mechanism.Methods SD rats were randomIy divided into four groups:sham operation group,modeI group,curcumin A group and curcumin B group,18 rats in each group.The rats of modeI group,curcumin A,B group of chronic cerebraI ischemia was prepared by the method of biIateraI carotid artery Iigation in rats,sham operation group and modeI group were given normaI saIine of the same.PI3K, AKT, protein expression were detected by ImmunohistochemicaI method,with the content of Caspase-3 determined by ELISA.The water content of brain tissue was detected by dry wet weight method, the permea-biIity of bIood brain barrier by methanamide method.Results Compared with modeI group,curcu-min group PI3K and AKT protein were significantIy increased (P〈0.05,P〈0.01),brain water content,caspase-3 and Evans bIue content were significantIy decreased ( P〈0.05,P〈0.01,P〈0.01).Conclusion The possibIe mechanism of the protective effect of curcumin on ischemic cerebraI injury is the penetration of the bIood-brain barrier after the activation of the PI3K/AKT signaIing pathway, and the expression of Caspase-3 mediated by it.
