摘要
探究超声辅助法提取紫花苜蓿多糖的最佳工艺,并对其进行体外抗肿瘤活性研究。利用超声辅助提取法提取紫花苜蓿多糖;利用细胞活力测定法、LDH活性测定和双荧光染色法研究紫花苜蓿多糖的体外抗肿瘤活性。细胞活力分析仪测定结果显示紫花苜蓿多糖能抑制人乳腺癌细胞的活性;LDH活性测定表明紫花苜蓿多糖能影响人乳腺癌细胞并呈现出剂量-效应关系;双荧光染色法结果表明紫花苜蓿多糖作用后人乳腺癌细胞凋亡数量较对照组增加;紫花苜蓿多糖具有较为良好的体外抗肿瘤活性。
To discuss the best ultrasonic-assisted extraction condition of the total polysaccharides from Medicago sativaL and to study the antitumor activity in vitro of total polysaccharides from Medicago sativaL. Ultrasonic-assisted extraction was employed in the extraction of the total polysaccharides from Medicago sativaL. Antitumor activity of total polysaccharides from Medicago sativaL was measured by using the cell viability assay, LDH activity and double staining immunofluorescence techniques. Cell viability assay and LDH activity measurement indicated that total polysaccharides from Medicago sativaL can dose-dependently inhibited the proliferation activity of human breast cancer cells. Double staining immunofluorescence techniques showed that human breast cancer cells apparently increased after the treatment of total polysaccharides from Medicago sativaL than the control groups. Total polysaccharides from Medicago sativaL had comparatively obvious antitumor activity in vitro.
