摘要
目的探讨糖尿病患者和糖尿病足溃疡患者外周血淋巴管内皮祖细胞的改变及其影响因素。方法采用RT-PCR检测外周血白细胞中淋巴管内皮祖细胞标志物VEGFR3mRNA表达;采用免疫组织化学检测外周血白细胞VEGFR3蛋白表达,并计数VEGFR3阳性细胞数量;检测各组患者临床指标,分别和外周血VEGFR3表达进行统计学相关分析。结果与正常对照组比较,实验组外周血VEGFR3mRNA表达及VEGFR3蛋白表达减少,差异有统计学意义(P<0.01)。糖尿病1组(Hb A1C<7%)与正常对照组比较,VEGFR3阳性细胞数量减少,差异有统计学意义(P<0.05);糖尿病2组(Hb A1C≥7%)和糖尿病足组与正常对照组比较,VEGFR3阳性细胞数量减少(P<0.01)。相关因素分析显示:VEGFR3表达的影响因素为糖化血红蛋白、空腹血糖、收缩压、舒张压。结论糖尿病及糖尿病足溃疡患者外周血淋巴管内皮祖细胞标志物VEGFR3表达减少,且和糖化血红蛋白、空腹血糖、收缩压、舒张压呈负相关。
Objective To explore changs of lymphatic endothelial progenitor cells and its affecting factorsin peripheral blood leukocytes of diabetes patients and diabetic foot ulcer patients. Methods The gene expression of VEGFR3 was detected by RT-PCR, the protein expression of VEGFR3 was determined by immuno-histochemistry, and the correlative factors were analyzed by statistical methods. Results Compared with normal control group,the gene and protein expression of VEGFR3 in diabetes group and in diabetic foot ulcer group were significantly deereased(P 〈 0.01 ). Number of positive cells of VEGFR3 in peripheral blood of diabetes group and diabetic foot ulcer group was decreased than that of normal control group (P 〈 0.01 ). The correlative analysis showed that factors decreasing VEGFR3 expression included HbA1C, systolic pressure, fasting blood-glucose and diastolic pressure. Conclusion The expression of VEGFR3 is decreased in peripheral blood of diabetes patients and diabetic foot ulcer patients, and there has negative correlation between VEGFR3 expression and HbA1C, systolic pressure ,fasting blood-glucose and diastolic pressure.
