新疆维吾尔族妇女宫颈鳞癌与人类白细胞抗原DQB1基因多态性的相关性分析

Correlation analysis of human leucocyte antigen-DQB1 allele polymorphism and cervical squamous carcinoma in Uygur women

摘要 目的 探讨新疆维吾尔族妇女宫颈鳞癌与人类白细胞抗原(HLA) DQB1基因多态性的相关性.方法 采用聚合酶链反应-直接测序分型法对80例于2009年8月至2010年4月在新疆维吾尔自治区人民医院就诊的新疆维吾尔族妇女宫颈鳞癌组织(观察组)及80例新疆维吾尔族妇女宫颈炎性组织(对照组)进行HLA-DQB1基因分型.比较2组HLA-DQB1等位基因及基因型频率,计算等位基因的比值比(OR)和95%置信区间(CI),用以估计等位基因与疾病联系的程度.结果 检测的160个样本中共含296个等位基因,其中杂合的等位基因型136人份,纯合的等位基因型24人份.经过频率计算和统计学分析发现,观察组HLA-DQB1* 0325(OR:10.60,95% CI:1.341~83.810)和HLA-DQB1* 0332(OR:12.59,95% CI:2.909~ 54.526)等位基因频率明显高于对照组,而HLA-DQB1女0317(OR:0.49,95% CI:0.304~0.798)和HLA-DQB1* 040302(OR:0.40,95% CI:0.243 ~0.658)等位基因频率明显低于对照组,差异均有统计学意义(均P<0.05).结论 新疆维吾尔族妇女宫颈鳞癌中HLA-DQB1基因型与我国其他地区报道有差异,等位基因HLA-DQB1*0325和HLA-DQB1* 0332可能是新疆维吾尔族妇女宫颈鳞癌的易感基因,而HLA-DQB1 * 0317和HLA-DQB1*040302可能是新疆维吾尔族妇女宫颈鳞癌的保护基因. Objective To investigate the correlation between human leucocyte antigen-DQB1 allele polymorphism and cervical squamous carcinoma in Uygur women.Methods The cervix tissue from 80 cases of cervical squamous carcinoma (observation group) and 80 cases of cervicitis (control group) from August 2009 to April 2010 were collected;polymerase chain reaction-sequencing based typing method was used to determine the HLA-DQB1 allele polymorphism.The allele type and frequency of allele types were compared between the two groups and the correlation between allele polymorphism with cervical squamous carcinoma was analyzed.Results Two hundred and ninety-six alleles were found in the 160 cases;among them 136 cases had heterozygous alleles and 24 cases had homozygous alleles.The frequencies of HLA-DQB1 * 0325 (OR:10.60,95% CI:1.341-83.810) and HLA-DQB1 * 0332 (OR:12.59,95% CI:2.909-54.526) in observation group were significantly higher than those in control group (P 〈 0.05),but HLA-DQB1 * 0317 (OR:0.49,95% CI:0.304-0.798) and HLA-DQB1 * 040302 (OR:0.40,95% CI:0.243-0.658) in observation group were lower thanthose in control group (P 〈 0.05).Conclusion The genotype of HLA-DQB1 in Uygur women is different from that in others;HLA-DQB1 * 0325 and HLA-DQB1 * 0332 may be the predisposing genes,meanwhile HLA-DQB1 *0317 and HLA-DQB1 * 040302 may be the protective genes.
