精浆游离MicroRNA 34-b在特发性少精子症及无精子症中的表达和临床应用 被引量:2

The expression and clinical role of seminal plasma free MicroRNA 34-b in patients with idiopathic oligospermia and azoospermia

摘要 目的研究游离Micro RNA 34-b(mi R-34b)在特发性少精子症和特发性无精子症患者精浆中的表达及诊断价值。方法收集特发性少精子症、特发性无精子症和同期因女方因素不孕前来检查的生育力评估正常的健康男性精液各20份,采用实时荧光定量PCR检测精浆游离mi R-34b的相对含量,通过方差分析统计各组间的差异,并对精浆游离mi R-34b在特发性少精子症和特发性无精子症中的诊断价值进行ROC曲线分析。结果正常对照组、特发性少精子症组和特发性无精子症组三组间精浆mi R-34b表达量差异有高度统计学意义(F=21.44,P〈0.01),与正常对照组相比,特发性少精子症组和特发性无精子症组精浆mi R-34b表达量显著降低,差异有高度统计学意义(P均〈0.01),但特发性少精子症组与特发性无精子症组间精浆mi R-34b相对含量差异无统计学意义(P=0.31)。ROC曲线显示mi R-34能够较好的区分特发性少精子症组与健康对照组以及特发性无精子症组与健康对照组曲线下面积分别为(AUC)为0.85(95%CI,0.73~0.97)、0.90(95%CI,0.80~0.99)。结论 mi R-34b在特发性少精子症和特发性无精子症患者精浆中明显降低,并对特发性少精子症和特发性无精症子症显示出一定的诊断价值。 Objective To quantify the expression of seminal plasma free mi R-34 b in patients with idiopathic oligospermia and azoospermia and to analyze the diagnostic value of it. Methods 20 cases semen of Idiopathic oligospermia and azoospermia were collected, respectively. At the same time, 20 cases semen of healthy male with fertility evaluation which because of the woman factor lead to infertility were collected as control group. the relative expression of seminal plasma free mi R-34 b was detected by real-time fluorescent quantitative PCR, the dates were analyzed by variance analysis, the diagnostic value of seminal plasma free mi R-34 b in diseases of oligospermia and azoospermia were analyzed by ROC curve. Results The relative expression of seminal plasma free mi R-34 b in oligospermia group and azoospermia group were decreased compared with normal controls(P〈0.01), while there were not change between oligospermia group and azoospermia group(P=0.31). ROC curve showed that mi R-34 b could distinguish between oligospermia disease and healthy control, the azoospermia and healthy control, the area under the curve were 0.85(95%CI,0.73-0.97) and 0.90(95% CI,0.80-0.99), respectively. Conclusion The expression of mi R-34 b was decreased obviously in patients with oligospermia and azoospermia,and shows certain diagnostic value.
