白藜芦醇对SO-RB50细胞裸鼠皮下移植瘤的作用效果

摘要 目的建立视网膜母细胞瘤细胞株SO-RB50皮下移植的裸鼠动物模型,观察白藜芦醇对肿瘤的影响。方法以2×108/ml SO-RB50细胞皮下注射制备视网膜母细胞瘤皮下移植动物模型。造模成功后将裸鼠随机分为实验组和对照组,实验组给予白藜芦醇、对照组给予等量生理盐水。测量裸鼠肿瘤体积并比较两组肿瘤重量;MTT法检测肿瘤细胞活性;蛋白印迹(Western blot)技术检测肿瘤增殖细胞核抗原(PCNA)、基质金属蛋白酶-2(MMP-2)、MMP-9的表达。结果与对照组比较,实验组肿瘤生长明显减慢,体积小于对照组(t=2.283,P<0.05);实验组肿瘤重量低于对照组,差异有统计学意义(t=4.535,P<0.05)。MTT结果显示实验组肿瘤细胞活性低于对照组,差异有统计学意义(t=8.764,P<0.05);Western blot结果显示,实验组PCNA、MMP-2、MMP-9蛋白表达均明显低于对照组,差异有统计学意义(t=5.812、7.077、6.216,P<0.05)。结论白藜芦醇可有效抑制SO-RB50细胞裸鼠皮下移植瘤的生长。 Objective Purpose of this study was to establish SO - RB50 cells transplanted model subcutaneously in nude mice, then effect of resveratrol on cancer cells was investigated. Methods The transplanted model was injected 2 × 10^8/mL SO - RB50 ceils subcutaneously in nude mice. The models were divided into experimen- tal group and control group randomly, experimental group were given resveratrol, and control group were trea- ted with normal saline. The volume of tumors was measured ; tumors weights of these 2 groups were compared ; cell activity of these 2 groups was tested with MTT assay ; expression of PCNA, MMP - 2, MMP - 9 proteins were tested with Western blot. Results The growth of tumors in experimental group was slower than that in control group ( t = 2. 283, P 〈 0. 05) . The average volume and weights of tumors in experimental group was smaller than that in control group ( t = 4. 535, P 〈 0. 05 ) . The tumor cell activity of experimental group was lower than that of control group (t =8. 764, P 〈0.05) . It was shown that expression of PCNA, MMP -2, MMP -9 proteins were lower in experimental group than that in control group (t=5. 812, 7. 077, 6. 216, P 〈 0. 05 ) . Conclusions Resveratrol could inhibit the growth of tumors in SO - RBSO cells transplanted model subcutaneously in nude mice.
