摘要
目的:研发一种快速、灵敏检测植物线虫的方法。方法:挑取单条线虫到裂解液中,用挑针压破线虫,以便提高蛋白质提取效率,利用MALDI-TOF质谱技术分析线虫的蛋白质指纹图谱,作为结果判断依据。结果:用该方法分析单条短体线虫的雌虫和二龄幼虫,雌虫的质谱峰值分别为m/z 2790、2970、3326、4373、4872、5654、7000、9579和19161,二龄幼虫的质谱峰值分别为m/z 2185、2790、3036、3326、4063、4373、4929、5380、6999和11156,二者相同的峰值分别为m/z 2790、4373和7000,其余峰值分别为各自的特征峰。结论:MALDI-TOF质谱能够区分同种线虫的不同虫态,检测灵敏度高,可用于单条植物线虫的检测。
Objective: To establish a rapid and sensitive method for detecting the plant nematode. Methods: A single nematode was picked into lysis liquid and crushed to improve the protein extraction efficiency. MALDI- TOF MS was used to analysis the protein spectra of single nematode. Results: The methode was used to analysis the single adult Pratylenchus spp and their second juvenile(J2) based on mass spectrometric profilling. Peaks at m/ z 2970, 3326, 4872, 5654, 9579, 19161 were only present in the adult, while m/z 2185, 3036, 3326, 4063, 4929, 5380, 11156 were unique to the J2. The common peaks to both the phases were at m/z 2790, 4373, 7000. Con- clusion: MALDI-TOF MS can be used to differentiate between the different stage of the nematode and the use of this method for efficient, sensitive and stable detection of a single nematode is feasible.
