摘要
目的:观察电针委中穴对大鼠腰椎间盘退变模型椎间盘组织中促炎因子IL-1β(interleukin-1beta)、基质金属蛋白酶-1(MMP-1)、基质金属酶组织抑制剂-1(TIMP-1)表达影响,探讨电针治疗腰椎退变疾病的可能起效机制。方法:将40只清洁级SD大鼠随机等分为正常组、模型组、电针委中组、电针非穴组共4组,采用纤维环穿刺法建立大鼠腰椎间盘退变模型。造模4周后,电针委中组、电针非穴组分别予相应电针干预,疗程结束后取出腰椎间盘组织行HE染色及RT-PCR检测IL-1β、MMP-1、TIMP-1基因表达水平。结果:与正常组相比,模型组、电针委中组、电针非穴组的IL-1β、MMP-1的mRNA表达升高, TIMP-1表达下降;与模型组相比,电针委中组及电针非穴组的IL-1β、MMP-1的mRNA表达下降,TIMP-1的mRNA表达升高;而电针委中组与电针非穴组比较,电针委中组的IL-1β、MMP-1的mRNA表达下降,TIMP-1的mRNA表达升高;各组间比较差异均有统计学意义(P〈0.05)。结论:电针可通过下调IL-1β、MMP-1的表达及上调TIMP-1的表达以延缓椎间盘退变,且电针委中穴刺激较非穴刺激效果更为显著。
Objective:Through the establishment of rat model of lumbar disc degeneration, observed in rat lumbar disc tissue morphological changes and the impact on mRNA expression of interleukin-1β(IL-1β), matrix metalloproteinase enzyme-1 (MMP-1) and tissue inhibitor of matrix metallo-proteinase enzyme-1 (TIMP-1) by electroacupuncture stimulation. To explore the electroacupuncture treatment of lumbar degenerative disease onset mechanism, for the clinical treatment of lumbar degenerative disease provide a theoretical basis by electroacupuncture. Methods: The 40 SD rats were randomly divided into four groups as normal group, model group, electroacupuncture stimulate“Weizhong”group, electroacupuncture stimu-late non-acupoint group. The normal group were routine to fed, and the remaining three groups establishment of rat model of lumbar disc degenera-tion by puncture the annulus. Four weeks after modeling, the model group did not interfere with electroacupuncture, the“Weizhong”group and the non-acupoint group electroacupuncture stimulated the corresponding parts. Each time treated 20min, once a day, ten times as a course of treatment, lasted three courses. After finishing treatment, the rats were sacrificed, lumbar intervertebral disc tissue were removed. Rat lumbar disc tissue mor-phological changes were observed by HE staining, and the expression levels of IL-1β, MMP-1, and TIMP-1 were applied by RT-PCR technology. Re-sult:On histological, the degree of lumbar disc degeneration from light to heavy followed normal group, electroacupuncture stimulate“Weizhong”group, electroacupuncture stimulate non-acupoint group, model group. Compared model group, electroacupuncture stimulate“Weizhong”group and electroacupuncture stimulate non-acupoint group with the normal group, the mRNA expression of IL-1Β,MMP-1 were increased, the mRNA expres-sion of TIMP-1 were decreased. Compared electroacupuncture stimulate“Weizhong”group and electroacupuncture stimulate non-acupoint group with the model group, the mRNA expression of IL-1Β, MMP-1 were decreased, the mRNA expression of TIMP-1 were increased. Compared elec-troacupuncture stimulate“WeiZhong”group with the electroacupuncture stimulate non-acupoint group, the mRNA expression of IL-1Β, MMP-1 were decreased, the mRNA expression of TIMP-1 were increased. Compare with a significant difference between the groups (P〈0.05). Conclusion:Electroacupuncture can reduce IL-1β, MMP-1, up-regulated TIMP-1, to delay the degeneration of intervertebral disc tissue. And electroacupuncture stimulation Weizhong acupoint stimulation than non-acupoint is more significant.
