BTLA及其负性调节慢性HBV感染的研究进展 被引量:2

摘要 淋巴细胞的活化依赖于双信号的共同刺激,其中第一信号由抗原特异性受体T细胞受体( TCR )或B细胞受体( BCR )与抗原结合产生;第二信号由共刺激分子或共抑制分子介导产生。共刺激分子和共抑制分子统称为共信号分子,分别行使正向激发作用和负向调节功能,两者发挥协同作用,共同维持机体的平衡[1]。共信号分子按照结构可分为两大家族:一类是免疫球蛋白( Ig )超家族(或称 B7/CD28超家族),包括 CD28、ICOS、CTLA-4、PD-1和BTLA( B and T lymphocyte attenuator );一类是肿瘤坏死家族( TNF )/TNF受体( TNFR )超家族[2],包括4-1 BB、CD27、CD30、HVEM 和 OX40。 BTLA 是继PD-1、CTLA-4后发现的第三个 CD28家族新成员[3],其配体并非B7家族成员,而是TNF家族的HVEM( Herpesvirus entry mediator ),打破了同种家族受体只与同一家族配体结合的观点。研究表明,与PD-1、CTLA-4一样, BTLA同样具有抑制T细胞反应及细胞因子产生的作用。在对乙肝病毒( HBV)感染的研究中也发现BTLA在病毒特异性T细胞上高表达,对T细胞增殖活性、分泌细胞因子功能等均具有强烈的抑制作用。现就BTLA及其配体的生物学特性及其在慢性HBV感染中的作用进行综述。
