摘要
目的探讨降钙素相关肽基因(calcitonin gene related peptide,CGRP)多态及其血清CGRP相关肽水平与1型糖尿病发生发展的关系。方法采用聚合酶链反应(polymerase chain reaction,PCR)技术,对42例1型糖尿病患者和126例正常对照的降钙素相关肽基因进行全长扩增并进行基因型分析,统计1型糖尿病组和正常对照组的CGRP主要的基因型基因频率,同时采用酶联免疫吸附法(enzyme-linked immunosorbent assay,ELISA)测定各组别各基因型血清降钙素基因相关肽浓度的差异。结果单变量Logistic回归分析显示类降钙素基因多态性(155209 C)与1型糖尿病发生的风险相关,相关系数为3.27,1型糖尿病组患者的血清CGRP浓度为正常对照组1.40倍。结论降钙素基因多态性与1型糖尿病遗传易感性相关,血清CGRP浓度可以作为1型糖尿病的新型标志物。
Objective:Discussion of β calcitonin gene(Human beta-CGRP gene exon 4 for calcitonin-like peptide,CGRP)the relationship between polymorphism and serum CGRP peptide level and the development of type 1 diabetes. Methods:Polymerase chain reaction(polymerase chain reaction,PCR),42 cases of type 1diabetic patients and 126 normal control class β calcitonin gene full-length amplification and genotyping,gene frequency statistics of type 1 diabetes group and normal control group beta calcitonin gene type,at the same time enzyme linked immunosorbent assay(enzyme-linked immunosorbent assay,ELISA)were determined by difference of calcitonin gene related peptide concentration in each category of each genotype serum. Results:Univariate Logistic regression analysis showed that beta calcitonin gene polymorphism(155209 C)are associated with the risk of type 1 diabetes,the correlation coefficients were 3.27,the concentration of serum CGRP of patients with type 1diabetes with normal control group 1.40 times. Conclusions:Calcitonin gene polymorphism and genetic susceptibility to type 1diabetes,serum CGRP concentration can be used as a novel marker for type 1 diabetes.
