体外受精-胚胎移植中卵丘颗粒细胞mtDNA4977bp缺失的研究 被引量:4

摘要 目的探讨高龄患者卵丘颗粒细胞mt DNA结构的完整性,为卵丘颗粒细胞mt DNA结构的完整性在辅助生殖技术中的应用提供理论依据。方法将接受体外受精-胚胎移植(IVF-ET)治疗的72例女性患者分为高龄组(年龄≥38岁)和非高龄组(年龄<38岁),收集其卵丘颗粒细胞,应用PCR方法检测mt DNA4977bp的缺失情况。结果高龄组患者的获卵数、优胚率及妊娠率均明显低于非高龄组(P<0.05)。两组患者均未检测到卵丘颗粒细胞mt DNA4977bp的缺失。结论接受IVF-ET治疗的女性患者,无论高龄与否,卵丘颗粒细胞mt DNA结构基本完整。 Objective:To research the structure integrity of Mitochondria DNA in cumulus granular cells of elderly patients,so as to provide theoretical evidence to the application of it in IVF-ET. Methods:72 women were divided into two groups:elder group(age≥38 years)and non-elder group(age〈38 years). Collected their cumulus granulosa cells,and detected the deletion of mitochondria DNA 4977 bp by PCR. Results:The number of oocytes retrieved,high quality embryo rate and pregnancy rate were obviously lower in elder patients than those in non-elder patients(P〈0.05). There was no deletion of mitochondria DNA4977 bp in the two groups. Conclusions:The structure of mitochondria DNA in cumulus granulosa cells of the patients whether elder or not is integrited in IVF-ET.
