淫羊藿苷上调miR-519d表达抑制人SKOV3卵巢癌细胞增殖、迁移和侵袭 被引量:7

Icariin inhibits proliferation, migration and invasion of SKOV3 ovarian cancer cells through upregulation of mi R-519d

摘要 目的淫羊藿苷(Icariin)对人SKOV3卵巢癌细胞增殖、迁移和侵袭的影响以及其可能的机制。方法针对SKOV3细胞,给予Icariin不同浓度处理不同时间后,采用CCK-8检测细胞增殖的变化;应用realtime PCR方法检测Icariin处理不同时间后mi R-519d在SKOV3细胞中的表达变化;利用Lipofect AMINE 2000将antimi R-519d以及其阴性对照质粒载体转染至SKOV3细胞,再给予Icariin,分别应用CCK-8法和Transwell小室法检测SKOV3细胞的增殖、迁移及侵袭能力变化。结果 Icarin显著抑制了SKOV3细胞的增殖,并且呈时间和剂量依赖;随时间变化,Icarin显著上调了mi R-519d在SKOV3细胞的表达水平;转染anti-mi R-519d后,Icarin的作用被阻断,SKOV3细胞的增殖,以及迁移、侵袭能力基本恢复。结论 mi R-519d参与了Icariin抑制SKOV3细胞的增殖,迁移和侵袭的调控过程。 Objective To study the effect and potential mechanism of Icariin on the proliferation, migration and invasion of human ovarian cancer SKOV3 cells. Methods The human SKOV3 ovarian cancer cells were cultured and treated with Icariin at different concentrations and different time, CCK-8 was used to detect the cell prolifration of SKOV3 cells, real-time PCR was used to detect the expression of miR-519 d in SKOV3 cells. Lipofect AMINE 2000 was used to transfect anti-miR-519 d plasmid vector into human SKOV3 cells, and then treated with Icariin. CCK-8 assay and transwell chamber assay were applied to measure the proliferation, migration and invasion of human SKOV3 cells. Results Icariin significantly inhibited the proliferation of SKOV3 cells in a time- and dose-dependent manner, and significantly increased the expression level of miR-519 d in SKOV3 cells in a time-dependent manner. The transfection of anti-miR-519 d blocked Icariin action, the proliferation, migration and invasion of SKOV3 cells recovered basically. Conclusion The upregulation of miR-519 d expression by Icariin inhibits the proliferation, migration and invasion of SKOV3 cells.
