摘要
将环介导等温扩增技术最初报道中的M13噬菌体引物排列成四种组合FIP+BIP+F3+B3、FIP+BIP+F3、FIP+BIP+B3、FIP+BIP,分别在不加M13噬菌体模板的情况下以相同的反应体系及反应条件进行等温扩增,琼脂糖凝胶电泳检测扩增产物,分析环介导等温扩增中产生假阳性的原因.结果表明,关于环介导等温扩增技术最初报道的试验具有非特异性扩增现象,环介导等温扩增的假阳性主要是引物二聚体引起的而并非是气溶胶污染导致的.
The primers for M13 phage in the initial report of Loop-mediated Isothermal Amplification (LAMP) were divided into 4 combinations: FIP + BIP + F3 + B3, FIP + BIP + F3, FIP + BIP + B3 and FIP + BIP, the isothermal amplification was performed with the same reaction system and reaction eonditions, respectively, no M13 phage DNA template was added to any of these reactions, the amplified products were deteeted with agarose gel eleetrophoresis, and the reasons of false positive occurred in LAMP were analyzed. The results indieated that the primers initially reported amplified non-specifically, the false positive amplification of LAMP was mainly due to primer dimers rather than aerosol pollution.
