摘要
目的探讨人乳头状瘤病毒(HPV)感染与宫颈癌组织中核转录因子κB(NF-κB)表达的关系,为进一步探索宫颈癌发生的原因以及研究相应的药物治疗提供试验依据。方法将医院2012年6月-2014年1月妇产门诊或住院收治的56例宫颈癌患者,同期收集对照组36例子宫肌瘤患者,采用免疫细胞化学染色法检测患者组织中NF-κB/p65、磷酸化核因子-κB(P-NF-κB/p65)蛋白的表达。结果 NF-κB/p65、P-NF-κB/p65的表达随宫颈癌病变的级别升高而升高,与宫颈癌病变级别呈正相关,差异有统计学意义(P<0.05);在NF-κB/p65、P-NF-κB/p65表达阳性的宫颈癌组织中,HPV16型阳性组的NF-κB表达的染色分值明显高于HPV16型阴性组,差异有统计学意义(P<0.05),提示HPV感染可以诱导NF-κB活化,NF-κB可能参与HPV16型的致癌过程,其表达随着宫颈病变的升高而升高,与宫颈病呈正相关;NF-κB/p65、P-NF-κB/p65在慢性宫颈炎组、CIN组和宫颈癌组的表达差异有统计学意义(χ2=24.39,P<0.05);NF-κB/p65、P-NF-κB/p65在宫颈癌的共表达差异有统计学意义(χ2=12.58,P<0.05)。结论人乳头状瘤病毒感染能促进NF-κB/p65、P-NF-κB/p65的表达和激活,NF-κB/p65、P-NF-κB/p65可能参与宫颈癌的发生和发展。
OBJECTIVE To investigate the relationship between Buman papillomavirus(HPV) type 16 infection and nuclear factor kappa B(NF‐κB) expression in cervical cancer ,so as to provide experimental basis for exploration of the causes of cervical cancer and the corresponding drug treatments .METHODS Totally 56 cases with cervical cancer in our hospital from Jun .2012 to Jan .2014 were collected and at the same time 36 cases of healthy people were collected as control group .Immunocytochemical staining method was adopted to detect the expression of NF‐κB/p65 and P‐NF‐κB/p65 protein .RESULTS The expression of NF‐κB/p65 and P‐NF‐κB/p65 increased with the increasing of the grade of cervical cancer ,and had positively correlation with the grade of cervical cancer .The difference was significant (P〈0 .05) .In cervical cancer tissues of positive expression of NF‐κB/p65 and P‐NF‐κB/p65 ,the expression of NF‐κB in HPV16 positive group was significantly higher than that in HPV16 negative group (P〈0 .05) ,which suggested that HPV infection can induce the activation of NF‐κB ,which may participate in HPV16 carcinogenic process and its expression will increase with the increasing of the grade of cervical cancer and had positively correlation with cervical disease .The differences of expression of NF‐κB/p65 and P‐NF‐κB/p65 in chronic cervicitis ,CIN group and cervical carcinoma group had significance (χ2 =24 .39 ,P〈0 .05) .The differ‐ences of co expression of NF‐κB/p65 and P‐NF‐κB/p65 in cervical cancer had significance (χ2 =12 .58 ,P〈0 .05) . CONCLUSION Human papillomavirus infection can promote the expression and activation of NF‐κB/p65 and P‐NF‐κB/p65 ,w hich may participate in the occurrence and development of cervical cancer .
