六味补气胶囊对肺气虚慢性阻塞性肺疾病大鼠模型肺组织代谢物及代谢特征标志物影响代谢组学随机平行对照研究 被引量:8

摘要 [目的]观察六味补气胶囊对肺气虚慢性阻塞性肺疾病大鼠模型肺组织代谢物影响及代谢特征标志物。[方法]使用随机平行对照方法,将56只雄性大鼠喂养40d,称重后使用随机数字表分为三组,空白对照组(24只),模型组(24只),干预组(8只)。参照"COPD肺气虚证动物模型"造模法,并复合木瓜蛋白酶雾化吸人法对模型组大鼠造模,造模期60d。灌胃干预。干预组六味补气胶囊(炙黄芪15g,人参、玉竹各10g,益智仁、陈皮各6g,肉桂3g),煎浓汁加水,配制成55%浓度,含生药量1.2g/m L,按1m L/100g体重,灌服2次/d,连续干预30d。观测大鼠肺功能。HPLC-MS/MS检测大鼠肺组织代谢物。[结果]模型检测:造模15d,肺功能、FVC、FEV0.3两组间无明显差异(P>0.05),FEV0.3/FVC模型组低于对照组(P<0.01);造模60d,模型组低于对照组(P<0.01)。肺功能检测:FVC、FEV0.3、FEV0.3/FVC模型组低于对照组(P<0.01),干预组高于模型组(P<0.01)。[结论]组胺、17-羟基亚麻酸、四氢叶酸酰-L-谷氨酰、前列腺素E2、乙烯乙酰氨基乙酸可能肺气虚型COPD大鼠模型疾病标志物-代谢特征。 [Objective]To observe the Fei-qi deficiency of chronic obstructive pulmonary disease rat model and the intervention of traditional Chinese medicine lung tissue metabolism.[Methods]Using a randomized parallel control method,56 male rats were fed 40d,after weighing using a random number table,the control group (24 rats),the model group (24 rats),the intervention group (8 rats).With reference to the patients of COPD with Fei-qi deficiency syndrome animal model modeling method,and papain nebulization ceiling law of model group rats model was made,the modeling for a period of 60 days.Irrigation intervention.Intervention group with Liuwei Buqi capsule (Astragalus root 15g,ginseng, polygonatum, 10g,Yizhiren tangerine peel,6g,cinnamon 3g),water fried gravy,formulated as a concentration of 55%,about per milliliter of liquid containing crude drug 1.2g,according to the weight of lmL/100g,irrigation service twice daily,30 days of continuous intervention.Observation of rat lung function.The metabolites of rat lung tissue were detected by HPLC-MS/MS technology.[Results]Model test:the lung function of rats in the two groups was detected by 15d,and there was no difference between the groups(P〉0.05),FEV0.3/FVC model group was lower than the control group (P〈0.01). The model group was lower than that of the control group (P〈0.01).Sixtieth days FEV0.3/FVC model group was lower than the control group (P〈0.01),the intervention group was higher than the model group (P〈0.01).Histamine,17- Hydroxylinolenic acid,Tetrahydrofolyl-[Glu](2),Prostaglandin,vinylace-tylglycine may be a marker for disease of COPD. [Conclusion]Get COPD rat model with lung Fei-qi deficiency type characteristic metabotites and find possible disease markers, traditional Chinese medicine in the treatment of metabolic characteristics have positive change.
