摘要
目的:通过观察HCN4通道在正常大鼠和膀胱过度活动症(OAB)模型大鼠膀胱中的表达情况,探讨HCN4通道在OAB中的作用及其与膀胱功能障碍的关系。方法:成功建立OAB大鼠模型后,将纳入实验的大鼠分为对照组、术后4周组、6周组及8周组,分别用免疫组织化学,Western blot检测OAB大鼠模型中HCN4通道的表达情况。结果:免疫组织化学结果与Western blot显示,HCN4通道在对照组、术后4周组、6周组、8周组各组间差异明显,组间分析结果显示强度最强的为8周组(P<0.05)。结论:OAB大鼠模型膀胱逼尿肌HCNC4通道表达较正常大鼠高,提示HCN4通道在OAB膀胱收缩功能的发病机制起重要作用,将来可能会作为OAB特异性药物治疗的新靶点。
Objective:To observe expression of HCN4 channel in both normal and overactive bladder(OAB)rats,to evaluate the influence of the HCN4 channel on OAB and to discuss the relationship between HCN4 and bladder dysfunction.Method:We divided the experimental rats into normal control group and OAB group(4w,6w,8w)after establishing the rats model of OAB successfully.By immunohistochemical detection and Western blot methods,we observed the expression of HCN4 in both normal rats and OAB rats.Result:This study indicated that HCN4 expression significantly increased in OAB rats compared with that in normal rats,especially in the8 wgroup by immunohistochemical detection and Western blot.Conclusion:Higher HCN4 expression in OAB rats than that in the normal rats is suggested that HCN4 play an important role in the mechanism of OAB.Maybe in the future it will be a new therapeutic strategy against OAB.
