HPLC法测定芎?汤活血有效部位中阿魏酸在大鼠体内药代动力学特征 被引量:1

摘要 目的:研究大鼠灌服芎汤活血有效部位后,有效成分阿魏酸在大鼠体内的药物代谢动力学规律。方法:大鼠灌服活血有效部位后,采集不同时间点的大鼠血样,采用HPLC法测定血样中阿魏酸的血药浓度并计算其药代动力学参数,建立药物代谢曲线。结果:阿魏酸在0.6132-81.76μg·mL^-1范围内线性关系良好,方法回收率均大于86%,日内日间精密度RSD均小于3%,其主要药动学参数分别为t1/2(37.8400±8.4090)min、Cmax(59.4200±17.9400)μg·mL^-1、AUC0-t(4815.7100±57.3400)μg·mL^-1·min^-1。结论:本试验建立了芎汤活血有效部位中有效成分阿魏酸在大鼠血浆中的HPLC测定方法,可用于大鼠血浆中阿魏酸血药浓度的测定及药动学研究。 Objective:To investigate the pharmacokinetics of ferulic acid in rats after administration of blood activating parts of XiongQiong Decoction. Methods:After administration of blood activating parts in XiongQiong Decoction,the plasma sample of rats were collected on different time points. The ferulic acid concentration in the plasma samples were detected by HPLC. The pharmacokinetic parameters were calculated and the metabolism curve was established. Results:Excellent liner relationship of ferulic acid was obtained in the range of 0.6132-81.76μg·mL^-1. The recovery of the method was more than 86% and the intra and inter day RSDs were all less than 3%. The pharmacokinetics parameters of ferulic acid were as follow:t1/2(37.8400±8.4090) min,Cmax(59.4200±17.9400)μg·mL^-1,AUC0-t(4815.7100±57.3400)μg·mL^-1·min^-1. Conclusion:The HPLC method for the determination of ferulic acid from Xiongqiong Decoction in plasma samples is established. It can be applied in the determination of blood concentration of ferulic acid in rats ’ plasma and pharmacokinetic study.
