摘要
目的观察低硒对大鼠心肌细胞内质网应激及细胞凋亡的影响。方法建立低硒仔三代大鼠(SDf3)、低硒仔三代补硒(SSf3)及对照组模型。采用颈动脉插管法测其心功能;Western blotting检测心肌组织中内质网应激标志蛋白GRP78、CHOP的表达变化。采用免疫组织化学SP法染色观察心肌细胞的凋亡情况。结果 SDf3组大鼠血硒及GPx活性明显低于对照组,而心肌组织GRP78、CHOP蛋白表达显著增高。SP法染色观察发现SDf3组大鼠心肌细胞凋亡明显增加(P<0.05)。结论低硒可以诱导大鼠心肌组织内内质网应激反应,内质网应激可能参与了心肌细胞的凋亡。
Objective To explore the effect of endoplasmic reticulum on cardiac myocyte apoptosis in selenium deficiency rats .Methods The selenium deficiency rats models with three generations ,selenium supplement group and control group were established respectively .The cardiac function of the rats was evaluated by carotid artery intubation .The protein expressions of GRP78 and CHOP were detected by Western blotting . The apoptosis of myocardial cells was detected by immunohistochemistry (SP method) .Results Serum selenium level and activity of GPx of blood were impaired in Se‐deficent rats .Expression of GRP78 and CHOP was significantly upregulated by treatment of Se deficiency . The number of apoptotic myocardial cells was increased in Se deficiency rats . Conclusion ER stress is involved in Se deficiency‐induced cardiac dysfunction .
