油茶皂苷与其脂质体体外溶血行为的对比研究

摘要 目的制备油茶皂苷(Sasanquasaponin,SQS)脂质体,对比考察SQS和SQS脂质体的体外溶血行为。方法薄膜分散-硫酸铵梯度法制备SQS脂质体。采用常规体外试管法(肉眼观察法)及改进的体外溶血性试验法(分光光度法),比较SQS和SQS脂质体体外溶血行为。结果体外溶血试验表明,SQS和SQS脂质体发生溶血浓度分别为4.40×10-5mg·m L-1和163.20×10-3mg·m L-1。结论 SQS脂质体明显减轻血管内溶血,减轻了SQS血管内溶血的不良反应,更安全有效。 OBJECTIVE Sasanquasaponin ( SQS SQS liposome in vitro were compared. METHODS ) liposome were prepared, the hemolytic activity of SQS and SQS liposome was made by film ultrasonic-ammonium sulfate gradient method. The hemocytolysis of SQS and SQS liposome were observed by means of the routine test tube method in vitro( macroscopic observation method)and modified hemolytic test method in vitro(spectrophotometry). The hemolytic activity was studied both in vitro. RESULTS Hemolytic crisis didn' t occur when the concentration of SQS ≤4. 40 × 10 ^-5 mg·mL^-1 and SQS liposome ≤163.20 × 10 ^- 3 mg · mL^-1, the hemolysis rate. CONCLUSION SQS liposome could reduce intravascular hemolysis inducedby puerarin, which is more safe, SQS liposome are optimistic to be used by intravenous injection.
