肺癌患者外周血Treg细胞与IL-10、TGF-β检测及其临床意义 被引量:20

摘要 目的测定肺癌患者外周血中CD_4^+CD_(25)^+FoxP3^+Treg细胞、CD_4^+CD_(25)^(hi)CD_(127)^(low)Treg细胞、IL-10及TGF-β的表达水平,并分析它们之间的相关性。方法采用流式细胞术检测73例肺癌患者外周血CD+4CD+25Fox P3+Treg细胞及CD_4^+CD_(25)^(hi)CD_(127)^(low)Treg细胞表达水平,采用酶联免疫吸附反应检测IL-10及TGF-β值,并与19例健康人进行比较。结果肺癌患者外周血CD+4CD+25Fox P3+Treg细胞、CD_4^+CD_(25)^(hi)CD_(127)^(low)Treg细胞、IL-10及TGF-β表达水平较正常对照组显著升高(P<0.05)。CD+4CD+25Fox P3+Treg细胞及CD_4^+CD_(25)^(hi)CD_(127)^(low)Treg细胞成正相关(P<0.05)。CD+4CD+25Fox P3+Treg细胞与IL-10、TGF-β成正相关(P<0.05)。CD_4^+CD_(25)^(hi)CD_(127)^(low)Treg细胞与IL-10、TGF-β成正相关(P<0.05)。结论CD_4^+CD_(25)^+FoxP3^+Treg、CD_4^+CD_(25)^(hi)CD_(127)^(low)这两种标记方法都是检测Treg细胞的可靠方式,Treg细胞、IL-10、TGF-β参与了肺癌患者肿瘤微环境的构成,是导致肺癌免疫功能抑制的原因之一,检测其水平变化可作为评估肺癌患者免疫功能的可靠指标。 Objective To observe the changes of the proportions of CD4+ CD2+5 FoxP3 + regulatory T cells, CD4+ CDhi25 CDlow127 regulatory T cells and the levels of IL-10 and TGF-βin peripheral blood of lung cancer. Methods The proportions of CD4+ CD2+5 FoxP3 + regulatory T cells, CD4+ CDhi25 CDlow127 regulatory T cells were determined by flow cytometry and the levels of L-10 and TGF-βwere measured with ELISA. 73 lung cancer patients were regarded as the experimental group and 19 health people as the control group. Results Compared with the control group, the propor-tions of CD4+ CD2+5 FoxP3 + regulatory T cells and CD4+ CDhi25 CDlow127 regulatory T cells and the levels of IL-10 and TGF-β increased significantly in the experiment group ( P〈0. 05 ) . CD4+ CD2+5 FoxP3 + regulatory T cells had a perfect positive correlation with CD4+ CDhi25 CDlow127 regulatory T cells and the levels of IL-10 and TGF-β(P〈0. 05). Conclu-sion CD4+ CD2+5 FoxP3 + gating method or CD4+ CDhi25 CDlow127 gating method can be used to detect Treg cells in humanperipheral blood. Treg cells, IL-10 and TGF-βparticipate in the composition of tumor micro-environment and are in-volved in immune suppression. Therefore, they can be used as indicators for evaluating lung cancer patients′immune functions.
