参麦注射液联用低分子肝素钙后在大鼠体内药动学特征研究 被引量:3

摘要 目的:以人参皂苷Rg1、Re、Rb1为参麦注射液药动学研究的指标成分,探讨低分子肝素钙对参麦注射液在大鼠体内药动学的影响。方法:采用HPLC方法测定大鼠血浆中人参皂苷Rg1、Re、Rb1的浓度,应用DAS 2.0药动学软件求算药动学参数。结果:参麦注射液与低分子肝素钙联用后人参皂苷Rg1、Rb1的血药浓度均升高,CL降低,MRT延长,AUC(0-144)和AUC(0-∞)均显著高于参麦注射液单独用药组;但联用低分子肝素钙后人参皂苷Re的血药浓度和药动学参数没有显著性变化。结论:本研究建立的测定大鼠血浆中人参皂苷Rg1、Re、Rb1的HPLC法,专属性强,精密度与回收率高,稳定性良好。首次探明低分子肝素钙对参麦注射液体内药动学的影响,为临床上中西药合理联用奠定研究基础。 Objective:Ginsenoside Rgl, Re, Rb1 were regarded as the major active constituents in Shenmai in- jection, to explore the effect of low molecular weight heparin on the pharmacokinetics of Shenmai injection. Methods: Concentrations of ginsenoside Rg1 , Re and Rb1 were measured in plasma by the established HPLC methods. The data obtained were processed by DAS 2.0 using non - compartmental pharmacokinetic analysis. Results : The plasma concentrations of ginsenoside Rg1 , Rb1 in rats were increased after Shenmai injection ad- ministration combined with low molecular weight heparin injection. CL was significantly lower;MRT was signif- icantly longer;AUC was significantly higher than that of Shenmai injection administration alone. Combined with low molecular weight heparin it did not affect the plasma concentrations of ginsenoside Re in rats, and there was no significant difference in the pharmacokinetie parameters of ginsenoside Re in rats. Conclusion: The specifity, precision, accuracy and stability are in line with the requirement of biological sample detection. The result proves the effect of low molecular weight heparin on the pharmacokinetics of Shenmai injection and provides reference for the combinnation with drugs for the treatment of diseases.
