雷马曲班对PAN诱导足细胞凋亡的影响

Effect of ramatroban on PAN-induced podocyte apoptosis

摘要 目的探讨血栓素A2受体(TXA2R)拮抗剂雷马曲班对嘌呤霉素氨基核苷(PAN)诱导足细胞凋亡的影响。方法条件永生性小鼠足细胞株分为A组(雷马曲班30nmol/ml)、B组(PAN 100μg/ml)和D组(PAN 100μg/ml+雷马曲班30nmol/ml),另设对照C组。流式细胞术检测细胞凋亡率,CCK-8法检测细胞增殖活性,RT-PCR和Western blot分别检测TXA2R mRNA和蛋白表达。结果与D、C组比较,B组足细胞凋亡率、TXA2R mRNA及蛋白水平均升高(P<0.01或P<0.05),增殖活性下降(P<0.01或P<0.05)。A组足细胞凋亡率、增殖活性、TXA2R mRNA及蛋白水平均与C组相仿(P>0.05)。结论雷马曲班对PAN诱导的足细胞凋亡具有抑制作用;其机制可能通过下调TXA2R表达而实现。 Objective To explore the effect of thromboxane A2 receptor(TXA2R) antagonist ramatroban on puromycin aminonucleoside (PAN )‐induced podocyte apoptosis .Methods The podocytes of conditionally immortalized mice were divided into groups of A (treated with ramatroban 30 nmol/ml) ,B(treated with PAN 100 μg/ml) ,D(treated with PAN 100 μg/ml plus ramatroban 30 nmol/ml) and C(without treatment as the control) .The apoptosis rate and activity of proliferation of podocytes were measured by flow cytometry and CCK‐8 method ,respectively .The expressions of TXA2R mRNA and protein were detected by RT‐PCR and Western blot ,respectively .Results The apoptosis rate and expression levels of TXA2R mRNA and protein were higher ,but the activity of proliferation was lower ,in group B than those in groups of D and C(P〈0 .01 or P〈0 .05) .Apoptosis rate ,proliferation activity and expression levels of TXA2R mRNA and protein in group A were similar to those in group C(P〉0 .05) .Conclusion Thromboxane has an inhibitory effect on PAN‐induced podocyte apoptosis ,possibly through down‐regulating TXA2R expression .
