摘要
目的探讨TNF-α、IL-6基因对高血糖"代谢记忆"的调控作用。方法根据正常对照(NC)组、糖尿病(DM)组、代谢记忆组对90只雄性健康SD大鼠进行造模,各30只。RT-PCR测定mRNA表达;TNF-α、IL-6的RT-PCR产物电泳条带积分光密度值(ROD);TNF-α、IL-6指标在血清中的表达情况。结果与NC组比较,代谢记忆组TNF-α、IL-6的RT-PCR产物ROD呈降低趋势,DM组呈升高趋势(P〈0.05);与NC组比较,代谢记忆组血清中TNF-α、IL-6水平呈降低趋势(P〈0.05),DM组呈升高趋势(P〈0.05)。结论 NC组TNF-α和IL-6的mRNA低于DM组,代谢记忆组(高血糖16~32mmol/L 12周在先,正常血糖5~8mmol/L 12周在后)的相应mRNA低于NC组。同上的规律在血清TNF-α和IL-6水平再次出现。这是临床糖尿病代谢记忆的分子生物学基础在TNF-α和IL-6的表现。
Objective To explore regulation of TNF-αand IL-6genes on ‘metabolic memory'of hyperglycemia. Methods A total of 90 healthy male SD rats were divided into three groups as control group(NC group,n=30),diabetes mellitus group(DM group,n=30),metabolic memory group(n=30).mRNA expression was measured by RT-PCR.Electrophoretic bands from TNF-αand IL-6 RT-PCR products were observed for integral optical density(ROD).Serum level of TNF-αand IL-6 were also tested. Results ROD of TNF-αand IL-6's RT-PCR products were decreased in metabolic memory group,increased in DM group as compared with NC group(P〈0.05).Serum level of TNF-αand IL-6were decreased in metabolic memory group,increased in DM group as compared with NC group(P〈0.05). Conclusion mRNA expression and serum level of TNF-αand IL-6is lower in NC group than in DM group,and also lower in metabolic memory group than in NC group.This finding illustrates the molecular biological mechanism of metabolic memory of diabetes mellitus by TNF-αand IL-6.
