摘要
为了节省水稻种子DNA提取时间,在超声波环境中进行CTAB温育前处理,并与常规环境温育处理的结果进行比较.结果表明,3种水稻样品颗粒细度与8种CTAB温育处理的24个组合中,超声波温育处理30与60 min对提取的DNA产量和水稻内源基因(GOS)的实时荧光PCR检测Ct值的影响,与相应同时间的常规温育处理比较,都达到差异显著水准,并达到常规温育8 h的效果,在确保检测结果准确的前提下,大幅度节省DNA提取时间,至少可节省7 h.
In order to save time in rice seed DNA extraction,CTAB incubating pretreatment was carried out in ultrasonic environment. The results showed that: in 24 combinations of three grades of rice grain fineness and eight kinds of CTAB buffer incubating treatments,the DNA yields and Ct value of real-time fluorescent PCR detection of endogenous genes( GOS) from ultrasonic incubating treatment for 30 and 60 min were significantly higher than that from conventional incubating treatment,which reached the effect of conventional treatment of 8h incubating with the time saving of 7 hours
