ALDH1表达对肝癌细胞株HepG2增殖的影响

摘要 目的探讨肝癌细胞株HepG2过表达ALDH1后对细胞增殖能力的影响。方法应用慢病毒稳定细胞株构建方法,分别构建pMSVC-空载和pMVSC-ALDH1细胞株。采用qRT-PCR法检测转染前后ALDH1mRNA表达的变化。采用MTT法检测HepG2细胞增殖的情况。结果成功建立转染pMVSC-ALDH1的肝癌细胞株,其ALDH1mRNA表达量明显增高。MTT法提示过表达ALDH1的HepG2细胞的增殖能力显著上升。结论ALDH1基因过表达可促进HepG2细胞增殖能力,ALDH1蛋白为肝癌治疗提供潜在靶点。
