应用Gibson Assembly法构建人果蝇zeste基因增强子同源物2基因过表达慢病毒载体

摘要 果蝇zeste基因增强子同源物2（EZH2）是组蛋白甲基转移酶。研究表明EZH2在前列腺癌中起关键作用[1]。我们应用GibsonAssembly法构建了pWPI-HA-EZH2慢病毒载体。 一、材料与方法 1．pWPI载体PacI和PmeI双酶切后，削平黏性末端，去磷酸化。 2．以pCMV-HA-EZH2质粒为模板，设计引物，序列：上游：CTCAGGAATTTCGACATTTTAAA佃IAATGGCATACCCATACGAT-GTTCCA，下游：TCCTGCAGCCCGTAGTTTTCAAGGGATITCCAT-TTCTCTTT。聚合酶链反应（PCR）扩增目的片段，回收产物。
