应用Gibson Assembly法构建人果蝇zeste基因增强子同源物2基因过表达慢病毒载体

摘要 果蝇zeste基因增强子同源物2(EZH2)是组蛋白甲基转移酶。研究表明EZH2在前列腺癌中起关键作用[1]。我们应用GibsonAssembly法构建了pWPI-HA-EZH2慢病毒载体。 一、材料与方法 1.pWPI载体PacI和PmeI双酶切后,削平黏性末端,去磷酸化。 2.以pCMV-HA-EZH2质粒为模板,设计引物,序列:上游:CTCAGGAATTTCGACATTTTAAA佃IAATGGCATACCCATACGAT-GTTCCA,下游:TCCTGCAGCCCGTAGTTTTCAAGGGATITCCAT-TTCTCTTT。聚合酶链反应(PCR)扩增目的片段,回收产物。
