摘要
目的:明确新疆地区水痘-带状疱疹病毒(VZV)流行的主要基因型。方法:采集6例水痘和119例带状疱疹患者的水疱液,提取标本中的DNA,PCR扩增ORF22、38、54、62 DNA片段并测序,测序结果与GenB ank中的VZV参考株基因序列进行比较,并用DNAMAN软件分析核酸序列和酶切位点。结果:PCR后电泳显示125份疱液标本中有120份VZV阳性。通过ORF22酶切位点分析,120份VZV临床分离株经序列对比J型79株,M1型40株,E型1株;通过ORF38、54酶切位点分析,119例VZV临床分离株为Pst I+Bgl I+型,1株VZV为Pst I+Bgl I-型;通过ORF62酶切位点分析,120株毒株均为Sma I-,都是野生株。结论:新疆地区水痘-带状疱疹病毒临床分离株主要为J型野生株。
{Abstract}: Objective:To genotype varicella-zoster virus ( VZV) strains isolated in Xinjiang. Methods:DNA was extracted from the blisters fluid in 6 patients with chickenpox and 119 patients with zoster. PCR and nucleotide sequencing were performed on the part of open reading frames ( ORF) 22, 38, 54 and 62. To con-firm the genotypes of isolated virus strains, the gene sequences obtained from PCR products were compared with that in GenBank of VZV strains, and the restriction enzyme sites were analyzed through DNAMAN soft-ware analysis. Results:One hundred and twenty samples of 125 were positive of VZV. Restriction enzyme site analysis of ORF 22 showed that 79 strains were genotype J, 40 were genotype M1 and 1 was genotype E. Re-striction enzyme site analysis of ORF38, 54 showed 119 strains were Pst I+Bgl I+, and 1 strain was Pst I+Bgl I-. Analysis of ORF 62 showed 120 strains were all wide-type Sma I-. Conclusion:Genotype J of the VZV i-solates is the major wild strain in Xinjiang region.
