摘要
目的:明确T细胞免疫球蛋白黏蛋白-3(Tim-3)对B16F10细胞共培养的小鼠脾淋巴细胞增殖的影响。方法:构建Tim-3真核表达载体后,CCK-8法检测Tim-3融合蛋白对B16F10共培养的小鼠脾淋巴细胞的增殖。结果:Tim-3融合蛋白转染组淋巴细胞增殖活力低于未转染组和转染空载体组,差异有统计学意义(P<0.05)。结论:Tim-3融合蛋白抑制B16F10共培养的小鼠脾淋巴细胞的增殖。
Objective:To determine the effect of T cell immunoglobulin mucin 3 ( Tim-3) on the prolifer-ation of mouse splenic lymphocytes co-cultured with B16F10 cells. Methods:After eukaryotic expression vec-tor was constructed successfully, the proliferation of splenic lymphocytes co-cultured with B16F10 cells was detected by Cell Counting Kit-8 ( CCK-8) method. Results:The proliferation activity of splenic lymphocytes co-cultured with B16F10 cells was lower in the trans-infection group than in the non-trans-infection and mock-vehicle groups ( P〈0.05) . Conclusion:Tim-3 can inhibite the proliferation of splenic lymphocytes co-cultured with B16F10 cells.
