摘要
为了研究食品浊度对叠氮溴化丙锭(PMA)-qPCR法检测食品中沙门杆菌活菌的影响,试验采用PMA—qPCR检测方法对不同浊度的食品进行分析。结果表明:当食品浊度小于23.4FTU时,经PMA处理后,能够实现对食品中沙门杆菌活菌的定量检测;当食品浊度为23.4~2.5×10^4FTU时,会干扰PMA处理效果,但可对食品中沙门杆菌活菌进行定性检测;当食品浊度大于2.5×10^4FTU时,干扰严重,PMA—qPCR法检测结果不可靠。通过增加PMA光照面积以及采用吸附柱法提取样品核酸,可提高PMA—qPCR法检测结果的准确性。
To study the effect of food turbidity on propidinm monoazide- quantitative PCR (PMA -qPCR) method for the detection of viable Salmonella spp. in food, PMA - qPCR detection method was used to analyze foods with different turbidity. The results showed that when the food turbidity was less than 23.4 FTU, it could realize the quantitative detection of viable Salmonella in food by qPCR after the PMA treatment ; when food turbidity ranged from 23.4 to 2.5 x10^4 FTU, the effect of PMA treatment was interfered, but it could achieve the qualitative detec- tion of viable Salmonella spp. in food ; when food turbidity was greater than 2.5x 10^4 FTU, the results of PMA - qPCR method were not relia- ble due to serious interference for the effect of PMA treatment. The accuracy of PMA - qPCR method for the detection of viable Salmonella spp. in food can be improved by increasing illumination area of PMA and extracting sample nucleic acid by adsorption column method.
