摘要
目的分离筛选出一株对甲胺磷敏感的双歧杆菌菌株。方法双歧杆菌菌株传代筛选,取一系列浓度(4.0、2.0、1.0、0.5和0.25μg/mL)的甲胺磷农药标准溶液30μL加入含TPY液体培养基的检测管溶液内,双歧杆菌菌株接种到检测管内,37℃厌氧培养12h,测定检测管液体吸光度(A)值。结果筛选一株甲胺磷敏感的短双歧杆菌菌株LJM-006,微生物抑制法的最佳接种浓度为107 CFU/mL,最低检测限为0.5mg/L。结论筛选出一株甲胺磷敏感的双歧杆菌LJM-006菌株,并可作为微生物法检测甲胺磷残留的菌株。
Objective To isolate and screen a Bifidobacterium strain with methamidophos sensitivity. Methods 30 μL of different concentration (4 μg/mL, 2μg/mL, 1 μg/mL, 0. 5 μg/mL, and 0. 25 μg/mL) of standard methami- dophos solution was added to the test tubes containing TYP liquid medium, respectively. Then Bifidobacterium strains were inoculated in the test tubes and cultured at 37 ℃ for 12 hours; the A value of the liquid in each test tube was detected. Results A strain of methamidophos-sensitive Bifidobacterium LJM-006 was obtained, the optimum inoculum concentration was 107 CFU/mL, and the detection limit was 0.5 mg/L with microbial in- hibition method. Conclusion A strain of methamidophos-sensitive Bifidobacterium LJM-006 was screen successfully, which can be used as the indicator bateria for detection of methamidophos.
