摘要
探讨霍夫变换(Hough transform)在聚合酶链反应-反向点杂交(polymerase chain reaction-reverse dot blotting,PCR-RDB)检测基因突变结果的自动化判读中的应用。以凯普HMM-2I医用核酸分子快速杂交仪检测基因突变为例,建立一种通过霍夫变换进行图像识别的方法,以快速判读PCR-RDB的检测结果,并提出一套基于移动终端的检测结果后处理方案。利用开发的软件,对113份地中海贫血常见突变以及86份人乳头瘤病毒分型的PCR-RDB检测结果进行了自动读取,并同人工判读结果进行了比较,二者的符合率均达100%。将霍夫变换应用于PCR-RDB检测图像的自动化判读符合实际需求,无需人工干预,可以降低肉眼判读的错误率并提高工作效率。
To explore the application of Hough transform for automated interpretation of the results of polymerase chain reaction-reverse dot blotting( PCR- RDB) for the detection of genetic mutations. The Hybribio HMM- 2I Rapid Hybridizer for the detection of DNA mutations was used as an example. A method was established for fast interpretation of the PCR- RDB results by image recognition using Hough transform. In addition,a set of mobile- terminals- based results processing scheme were proposed. A software was developed for automatic interpretation of the PCR- RDB results for detecting common mutations in 113 thalassemia cases and subtypes of human papilloma virus in 83 cases. The results were compared with those of manual interpretation,with a consistency of 100%. The Hough transform can suit automated interpretation of the results of PCR- RDB assays. The method does not require human intervention,can reduce the error rate of visual interpretation,and improve the efficiency of testing.
