Th1和Th2细胞来源的儿茶酚胺在胶原诱导性关节炎中的作用

摘要 目的探讨Th1和Th2细胞来源的儿茶酚胺在胶原诱导性关节炎(CIA)疾病发生、发展过程中的作用。方法 18只DBA/1小鼠随机均分为对照组(A组)、35dCIA模型组(B组)和55dCIA模型组(C组)。采用实时定量PCR检测T-bet、IFN-γ、IL-2、GATA-3、IL-10和IL-4mRNA表达,免疫荧光染色观察淋巴结中酪氨酸羟化酶、囊泡单胺转运体2、单胺氧化酶分别与T-bet和GATA-3的共定位情况。结果与A组相比,B组和C组小鼠淋巴结中T-bet、IFN-γ和IL-2mRNA表达增加,IL-10和IL-4 mRNA表达减少,Th1细胞合成儿茶酚胺的能力增强(P<0.05)。结论 CIA小鼠淋巴结中Th1细胞来源的儿茶酚胺增加;其参与了CIA发生、发展过程,并发挥抗炎作用。 Objective To investigate the effect of Th1 and Th2 cells-derived catecholamines on the development and progression of collagen-induced arthritis(CIA) .Methods Eighteen DBA/1 mice were equally randomized into three groups of A(normal control) ,B(CIA model established on the 35th day) and C(CIA model established on the 55th day) .The mRNA expressions of T-bet ,IFN-γ,IL-2 , GATA-3 ,IL-10 and IL-4 were detected by real-time PCR .The colocalization of tyrosine hydroxylase (TH) ,vesicular monoamine transporter-2(VMAT-2) and monoamine oxidase(MAO) with T-bet and GATA-3 in lymph nodes was observed by immunofluorescent staining .Results Compared with group A ,the mRNA expressions of T-bet ,IFN-γ and IL-2 were increased ,mRNA expressions of IL-10 and IL-4 were decreased ,and ability of catecholamine synthesis in Th1 cells was enhanced in groups of B and C ( P〈0 .05 ) .Conclusion Th1 cells-derived catecholamines in lymph nodes of CIA mice are increased ,which may play an anti-inflammatory role in the development and progression of CIA .
