摘要
目的研究系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMCs)Nr4a1基因m RNA表达水平。方法通过Ficoll梯度离心法分离出47例SLE患者和30例健康人PBMCs,用TRIzol提取总RNA,再将总RNA逆转录成c DNA,以荧光定量PCR法检测Nr4a1基因m RNA水平,比较SLE患者组和健康对照组的Nr4a1基因m RNA水平差别,分析SLE患者Nr4a1基因m RNA水平与补体C3、补体C4、SLE疾病活动度指数(SLEDAI)的相关关系。结果与健康对照组比较,SLE患者组PBMCs中Nr4a1基因m RNA水平明显降低,差异有统计学意义。结论 SLE患者PBMCs中Nr4a1基因m RNA水平显著低于健康人。
In order to investigate Nr4a1 m RNA expression level in peripheral blood mononuclear cells(PBMCs) of patients with systemic lupus erythematosus(SLE), we separated PBMCs from 47 SLE patients and 30 healthy control people by Ficoll gradiant centrifugation. Total RNA, which had been isolated using TRIzol, was reverse-transcribed into c DNA, and then the m RNA expression level of gene Nr4a1 was detected through real-time fluorescent quantitative PCR. The Nr4a1 m RNA expression level of SLE patients was compared with that of healthy control people; the correlation of Nr4a1 m RNA level with complement 3(C3), complement 4(C4), SLE disease activity index(SLEDAI) was analyzed. Data showed that, when compared with healthy subjects, Nr4a1 m RNA expression level of SLE patients was lower with statistically significance. In conclusion, Nr4a1 m RNA expression level in PBMCs of SLE patients is significantly lower than that of healthy subjects.
